Short-chain fatty acids induced autophagy serves as an adaptive strategy for retarding mitochondria-mediated apoptotic cell death

Abstract: Short-chain fatty acids (SCFAs) are the major by-products of bacterial fermentation of undigested dietary fibers in the large intestine. SCFAs, mostly propionate and butyrate, inhibit proliferation and induce apoptosis in colon cancer cells, but clinical trials had mixed results regarding the anti-tumor activities of SCFAs. Herein we demonstrate that propionate and butyrate induced autophagy in human colon cancer cells to dampen apoptosis whereas inhibition of autophagy potentiated SCFA induced apoptosis. Colo… Show more

“…25 Additionally, in cancer cells, butyrate treatment induced autophagy as a defense strategy to retard mitochondrial apoptosisinduced cell death. 26 We observed lower apoptosis levels and higher levels of the ATG-7 protein after acetate treatment, suggesting that acetate could induce autophagy and thereby retard apoptosis.…”

“…Under chronic or irreversible ER stress, cells undergo apoptosis, where the Bcl-2 protein family plays a crucial role at the mitochondria to trigger cytochrome c release and formation of apoptosome (Rodriguez et al 2011). The ability of ABT-737 (Malik et al 2011) as well as HDACi (Shao et al 2004;Tang et al 2011) to induce autophagy has already been documented. Our results could be in favour of inducing autophagy by NaBu.…”

“…In addition, another short-chain fatty acid, propionate, was shown to inhibit HDAC activity (Davie 2003). Both compounds induce autophagy with the up-regulation of LAM-2 (lysosomal-associated membrane protein 2) expression and LC3 conversion (Tang et al 2011). However, considering the low stability of this compound, its use is limited to in vitro studies.…”
