Shoot Development through Modified Transverse Thin Cell Layer (tTCL) Culture of Phalaenopsis Hybrid Protocorms

Abstract: This first-attempt study used microtome-based methods to generate a thin cell layer culture for the micropropagation of Phal. Hwafeng Redjewel × Phal. New Cinderella. Protocorms were embedded in various agarose concentrations (8–12%, w/v) and dried from 1 to 8 h before sectioning with a microtome. Optimal conditions for slicing sections of 100 to 300 μm were achieved when the protocorms were embedded at 10% (w/v) agarose and dried for 4 h under laminar flow, and the hardness of the agarose block under these co… Show more

“…The similar finding of inhibitory effect of BAP on the conversion of protocorms into plantlets and root formation was reported in Dendrobium nobile (Nayak et al 2002), Dendrobium huoshanense (Luo et al 2009) and Doritis pulcherrima (Mondal et al 2013). In contrast to these results BAP also stimulated the formation of highest number of shoots in Dendrobium chryseum (Maharjan et al 2020), highest percentage of callus in Dendrobium barbatulum (Ashok Pyati, 2020) and highest mean values of PLBs in Phalaenopsis hybrid (Lo et al 2022). However, NAA was showed inhibitory effect to induce either EC or SEs in the medium.…”

“…There are reports in which material was harvested from aseptic mother stock maintained on basal medium and TCL sections were aseptically excised (Bacopa monnieri [10]; Hadrolaelia grandis [11]; Paphiopedilum callosum [12]; Dendrobium spp. [13]; Begonia tuberous [14]; orchid Phalaenopsis hybrid protocorms [15]; Malus spp. sprouts [16,17]).…”

“…In many studies, protocorms were used for propagation and production of seedlings for several genera of orchids such as Phalaenopsis and Cymbidium (Lo et al, 2022;Regmi et al, 2017). This can be explained by the fact that protocorm cells have the ability to divide, which makes them ideal explants in the process of clonal micropropagation and studying the morphology of growth and development of representatives of the orchid family (Chookoh et al, 2019;Yeung, 2017).…”
