Shikonin inhibits maturation of bone marrow‐derived dendritic cells and suppresses allergic airway inflammation in a murine model of asthma

Abstract: BACKGROUND AND PURPOSEShikonin exhibits a wide range of anti-inflammatory actions. Here, we assessed its effects on maturation of murine bone marrow-derived dendritic cells (BM-DCs) and on allergic reactions in a murine model of asthma. EXPERIMENTAL APPROACHCultured murine BM-DCs were used to investigate the effects of shikonin on expression of cell surface markers and their stimulation of T-cell proliferation and cytokine production. The therapeutic potential of shikonin was evaluated in a model of allergic a… Show more

“…Mice were given an aerosol challenge of 5 mL 4% OVA in PBS once a day, 5 days per week, for 4 weeks. On days 29,31,33,36,38,40,43,45,47,50,52, and 54, mice were given an intratracheal instillation of PBS (OVA group), with or without anti-HMGB1 neutralizing antibody (30 mg/mouse; OVA + anti-HMGB1 Ab group), or a control rabbit IgG antibody (Jackson ImmunoResearch Laboratories Inc., West Grove, PA, USA) (30 mg/mouse; OVA + isotype control Ab group) 1 h before OVA exposure. There were 8-10 mice per group.…”

“…The dyes and related solutions purchased from Sigma. Quantification of airway inflammation, PAS-positive bronchial epithelial cells, and collagen fibril deposition were performed according to methods reported in a previous study [31,32]. For immunohistochemistry, after deparaffinization, lung sections were hydrated, and epitope retrieval was performed according to the manufacturer's protocol (DakoCytomation Target Retrieval Solution, pH 6.0; DakoCytomation, Carpinteria, CA, USA).…”

“…(Boraginaceae) (Brigham et al, 1999;Yang et al, 2009). Shikonin and its analogues are potential pharmaceutical agents with anticancer activities in addition to anti-inflammatory properties (Lee et al, 2010;Wang et al, 2013;Yao & Zhou., 2010). Shikonin preferentially inhibits the growth of human epidermoid carcinoma cells in a concentration-and time-dependent manner (Singh et al, 2003).…”
