Shh signaling is essential for rugae morphogenesis in mice

Abstract: Palatal ridges, or rugae palatinae, are corrugated structures observed in the hard palate region. They are found in most mammalian species, but their number and arrangement are species-specific. Nine palatal rugae are found in the mouse secondary palate. Previous studies have shown that epithelial Shh signaling in the palatal ridge plays an important role during rugae development. Moreover, Wnt family members, including LEF1, play a functional role in orofacial morphogenesis. To explore the function of Shh dur… Show more

“…, 30 Orientation of the primary palatal rugae was found to have significant differences on the left side, and the first primary rugae were found to be posteriorly directed more frequently in Class II division 2 group. Previous study reported insignificant differences in the orientation of primary palatal rugae 7 . The possible reason of conflicting results could be due to the fact that rugae are asymmetric structures and the mechanism of their development and establishment has been poorly understood 31 .…”

“…Scientific Reports | (2020) 10:19074 | https://doi.org/10.1038/s41598-020-76240-w www.nature.com/scientificreports/ broader and ectopic rugae distributed in a widespread manner across the palate 23,56 ; whilst conditional loss of Shh in the oral epithelium or pharmacological inhibition in palatal shelf explants also results in the formation of highly disorganised and ectopic rugae 23,57 . Together, these data suggest that much like Sprouty, SHH acts as an inhibitor within this system.…”

“…The Shh pathway is also implicated in patterning cusps and teeth. Extra incisors and molars and abnormal rugae patterns were reported in mice with suppressed Shh activity (Cho et al 2011; Lee et al 2011). Suppression of Shh signaling activity in vitro increases the number of cusps in wild-type tooth germs.…”
