SHARP1 Suppresses Angiogenesis of Endometrial Cancer by Decreasing Hypoxia-Inducible Factor-1α Level

Abstract: Recent data support a role for SHARP1, a basic helix-loop-helix transcription repressor, in the regulation of malignant cell behavior in several human cancers. However, the expression and role of SHARP1 during the development of endometrial cancer (EC) remain unclear. Here we show that upregulation of SHARP1 suppressed tumor angiogenesis by decreasing hypoxia-inducible factor-1α (HIF-1α), inhibited cell viability and tumor growth in EC. Immunohistochemical staining showed that the expression of SHARP1 was nega… Show more

“…Tube formation assay. We coated a 96-well plate with 100 µl Matrigel per well and incubated the plate at 37˚C for 2 h. Then, 2x10 4 HUVECs were suspended in 1 ml diluted HEC-1B CM (dilution in 1:10) and applied to the pre-coated 96-well plate at a density of 2000 cells/well (19). After incubation at 37˚C Diego, CA, USA).…”

“…Endometrial carcinoma (EC) is a significant cause of morbidity and mortality for women across the world [1]. EC in its early stages is often satisfactorily treated with surgery, but advanced EC is characterized by difficult treatment and a poor prognosis [2], especially in terms of recurrent or metastatic disease [3].…”

“…Angiogenesis refers to the formation of novel blood vessels from preexisting ones and plays a major role in tumor growth, development and progression, and metastasis [1]. Vascular endothelial growth factor A (VEGFA) is a pro-angiogenic factor involved in the proliferation, differentiation, and migration of endothelial cells and participates in vascular permeability [19].…”
