Shared Genetic Liability and Causal Associations Between Major Depressive Disorder and Cardiovascular Diseases

Abstract: Major depressive disorder (MDD) is phenotypically associated with cardiovascular diseases (CVD). We aim to investigate mechanisms underlying relationships between MDD and CVD in the context of shared genetic variations. Polygenic overlap analysis was used to test genetic correlation and to analyze shared genetic variations between MDD and seven cardiovascular outcomes (coronary artery disease (CAD), heart failure, atrial fibrillation, stroke, systolic blood pressure, diastolic blood pressure, and pulse pressur… Show more

“…Mendelian randomization (MR) is an analytic framework that utilizes genetic variants as instrumental variables to test the causative association between an exposure and an outcome ( 19 ), which has been widely used in recent studies ( 20 22 ). In particular, previous MR analyses have reported causal risk factors for a severe course of COVID-19, including body mass index and smoking intensity ( 23 25 ).…”

“…Third, though handgrip strength is an objective and common marker of muscular strength, it mainly represents upper body strength. Fourth, as the causal association was evaluated using MR based on the genetic information of each trait, the result should be interpreted with caution ( 42 ), with the understanding that the development of handgrip strength and CVDs were multifactorial and involved interactions among plenty of psycho-social-environmental factors ( 43 ). Finally, some samples in the GWAS of AF and HF have also been included in the UK Biobank, which may introduce bias.…”

“…First, although we have excluded potential pleiotropic SNPs and MR-Egger regression indicated little horizontal pleiotropy, the influence of pleiotropy bias remains a concern in this study. Specifically, psychiatric traits such as major depressive disorder and neuroticism may be confounding factors in the causal associations between asthma and CVDs ( Cao et al, 2021 ; Wu et al, 2021 ; Zhang et al, 2021 , 2022 ). Besides, the divergent results of sensitivity analyses may indicate a bias of genetic pleiotropy towards asthma-AF and asthma-HF associations that we observed.…”
