sFRP2 Suppression of Bone Morphogenic Protein (BMP) and Wnt Signaling Mediates Mesenchymal Stem Cell (MSC) Self-renewal Promoting Engraftment and Myocardial Repair

Abstract: Transplantation of mesenchymal stem cells (MSCs) is a promising therapy for ischemic injury; however, inadequate survival of implanted cells in host tissue is a substantial impediment in the progress of cellular therapy. Secreted Frizzled-related protein 2 (sFRP2) has recently been highlighted as a key mediator of MSC-driven myocardial and wound repair. Notably, sFRP2 mediates significant enhancement of MSC engraftment in vivo. We hypothesized that sFRP2 improves MSC engraftment by modulating self-renewal thro… Show more

“…The Wnt signaling pathway influences cell-cell communication, adult tissue maintenance, and gene expression. BMPR2 signaling may regulate both canonical and noncanonical Wnt pathways in EC and MC to influence proliferation, survival, and motility during angiogenesis and remodeling of the pulmonary circulation (3,19,39). While the relationship of BMPR2 and Wnt pathways has been defined during development, the regulatory targets of Wnt signaling, common across multiple cell types, in BMPR2-associated PAH are unknown.…”

“…Mesenchymal stem cells (MSCs) have extensive self-renewal potential, can modulate immunocyte activation [1,2], are easily expanded and stored ex vivo, and are considered to be ''immunoprivileged cells.'' Under stress conditions, MSCs decrease local inflammation and participate in tissue repair and regeneration by secreting cytokines, chemokines, and extracellular matrix proteins [3,4].…”

“…In addition to its use in studying a wide array of attributes of wound healing, the PVA sponge has also been used in many other types of studies. It has been utilized to investigate tumor angiogenesis, drug delivery and stem cell survival and engraftment 1,2,6,7 . With its great alterability, prior extensive use, and reproducible results, the PVA sponge is an ideal model for many studies 1,2 .…”
