Sexing Frogs by Real-Time PCR: Using Aromatase (cyp19) as an Early Ovarian Differentiation Marker

Abstract: Most anurans have no identified sex-markers; therefore, alternative methods for identification of early changes in sex ratios are required. In this study, Lithobates sylvaticus and Silurana tropicalis tadpoles were sampled at different developmental stages covering the entire process of sex differentiation. Three candidate genes known to be involved in sex differentiation in other vertebrate species were selected to develop a method to identify phenotypic sex in frogs: cytochrome p450 aromatase (cyp19), forkhe… Show more

“…Gonadal development and differentiation in anurans starts between Gs28 and Gs31 [Ogielska and Kotusz, 2004]. Our study did not detect sex differences in star at the onset of gonadal differentiation (Gs30), a stage where we have previously demonstrated clear sexually dimorphic expression of genes involved in gonadal differentiation in L. sylvaticus tadpoles [Navarro-Martín et al, 2012]. These results are in accordance with Maruo et al [2008], who examined star mRNA levels at different developmental stages during premetamorphosis in G. rugosa (corresponding to developmental stages between Gs25 and Gs30).…”

“…Tadpoles in this particular pond underwent metamorphosis by mid-July, and the pond dried completely at the end of August. In a previous study the samples were used to develop a PCR-based method to predict phenotypic sex in tadpoles of different amphibian species [Navarro-Martín et al, 2012]. To do that, we measured tadpoles' mRNA levels of candidate genes known to be involved in sex differentiation in other vertebrate species.…”

“…Particularly, the expression of aromatase is higher in the undifferentiated gonad of female tadpoles than in male ones, while CYP17 is enhanced in the undifferentiated male gonads prior to sex determination and continues at high levels thereafter (Sakurai et al, ; Nakamura, ). This same pattern was found in X. laevis and Lithobates sylvaticus (Okada et al, ; Navarro‐Martín et al, ).…”

“…This increasing expression is correlated with ovarian formation [Okada et al, 2009]. Similarly, in the adult wood frog, Lithobates sylvaticus , Foxl2 is expressed in both gonads although a clear dimorphic expression in the ovaries is observed [Navarro- Martin et al, 2012]. Hence, in amphibians Foxl2 is also linked to gonad feminization.…”
