Sex-specific and opposite modulatory aspects revealed by PPI network and pathway analysis of ischemic stroke in humans

Abstract: BackgroundIschemic Stroke (IS) is a major disease which greatly threatens human health. Recent studies showed sex-specific outcomes and mechanisms of cerebral ischemic stroke. This study aimed to identify the key changes of gene expression between male and female IS in humans. MethodsGene expression dataset GSE22255, including peripheral blood samples, was downloaded from the Gene Expression Omnibus (GEO) dataset. Differentially Expressed Genes (DEGs) with a LogFC>1, and a P-value <0.05 were screened by BioCon… Show more

“…In static PPI network analysis, several thresholds of STRING combined scores (0.4, 0.7, and 0.9) are routinely considered as a cut‐off value. In this regard, more authentic PPI studies often use interactions with the combined scores of 0.4 and more to identify significant modules; 63–66 while others use 0.7 and 0.9 as a significant cut‐off value 67,68 . To solve this discrepancy, altering the expression of several genes with different combined scores was investigated in response to RAB5A knockdown.…”

“…CXCL9 was the most prominent gene, and was regulated by lncRNA RP11‐1094M14.8 and miR‐1269a (Figure 3A). Based on the ceRNA network, mRNA interaction relationships were identified using the STRING database with scores of >0.4, and 60 mRNAs were selected to construct the PPI network 31 . CXCL9 also showed important regulatory effects in the PPI network (Figure 3B).…”

“…The progression of stroke is affected by the intricate relationship between the blood-brain barrier (BBB) and TNF (Pan and Kastin, 2007). A previous protein-protein interaction (PPI) network-based analysis also revealed the involvement of the TNF signaling pathway in stroke (Lv et al, 2020). Furthermore, a TCM prescription, Huang-Lian-Jie-Du Decoction, has 28 target proteins in stroke network, while it alleviates stroke via a pharmacological mechanism in part by mediating the TNF signaling pathway (Wang et al, 2019).…”
