Sex-Biased Gene Expression and Isoform Profile of Brine Shrimp Artemia franciscana by Transcriptome Analysis

Abstract: The brine shrimp Artemia has a ZW sex determination system with ZW chromosomes in females and ZZ chromosomes in males. Artemia has been considered a promising model organism for ZW sex-determining systems, but the genes involved in sex determination and differentiation of Artemia have not yet been identified. Here, we conducted transcriptome sequencing of female and male A. franciscana using PacBio Iso-Seq and Illumina RNA-Seq techniques to identify candidate sex determination genes. Among the 42,566 transcrip… Show more

“…Two different A. franciscana de novo transcriptome assemblies were made using Trinity. The first using pooled RNA-seq reads from male heads and testes [2 replicates each ( Huylmans et al 2019 )], and the second using the published whole-body male RNA-seq library (SRR14598203, Jo, Lee, Choi, Kim, Lee, and Park 2021 ). The 2 assemblies were concatenated and then filtered using the tr2aacds.pl script from EvidentialGene, and mapped to the A. sinica genome using the same procedure as described in section Mapping of W candidates to the A. sinica genome .…”

“…Eight sex-linked AFLP marker alleles have been reported in A. franciscana that are inherited from the female parent [ 1 ]. Some genes related to sexual differentiation in A. franciscana have been identified and shown as sex-biased gene expression [ 8 ]. Masculinizer gene ( Ar-Masc) was identified in A. franciscana [ 9 ], the MASC protein encoded in the Z chromosome activates a sex-determining gene that regulates gene splicing of Doublesex ( dsx ) into the male-specific isoform in the silkworm Bombyx mori [ 10 ].…”
