Severe CD4 T Cell-Mediated Immunopathology in Murine Schistosomiasis Is Dependent on IL-12p40 and Correlates with High Levels of IL-17

Abstract: C57BL/6 mice infected with the helminth Schistosoma mansoni develop small hepatic granulomas around parasite eggs, but concomitant immunization with soluble schistosome egg Ags (SEA) in CFA (SEA/CFA) causes marked exacerbation of the lesions in a Th1-dominated environment characterized by high levels of IFN-γ. We explored the cause of the severe immunopathology by using IL-12p40−/− and IL-12p35−/− mice. SEA/CFA-immunized IL-12p40−/− mice, incapable of making IL-12 or IL-23, were completely resistant to high pa… Show more

“…However, the involvement of IL-17A in granuloma formation is not restricted to bacterial infection. The infection of mice with Schistosoma mansoni postimmunization with S. mansoni egg Ag induces severe granulomas, and the immunopathology depended on IL-17A (48). In contrast to mycobacterial infection, the IL-17A-producing cells were S. mansoni Ag-specific T cells.…”

“…Mice infected with Schistosoma mansoni and then immunized with soluble schistosome egg antigens (SEA) in CFA develop severe egg-induced immunopathology, with granulomatous inflammation mediated by IL-17 [97]. Furthermore S. mansoni-infected IL-23p19 À/À mice with or mice treated with anti-IL-17 neutralizing antibody had significantly reduced hepatic granulomatous inflammation, suggesting that IL-17-producing T cells contribute to immunopathology [84].…”

“…IL-17A and IL-17F are important members of the IL-17 cytokine family preferentially produced by helper T 17 (Th17) cells, which are responsible for the pathogenic activity of the lineage of CD4+ effector cells and multiple proinflammatory mediators (Rutitzky et al, 2005;Ishigame et al, 2009 et al, 2010). However, subsequent replication studies for the association between IL-17A and IL-17F variants and gastric cancer risk are controversial (Shibata et al, 2009;Arisawa et al, 2012;Rafiei et al, 2013;Zhang et al, 2014;Zhu et al, 2014).…”
