Sestrin2 Signaling Pathway Regulates Podocyte Biology and Protects against Diabetic Nephropathy

Abstract: Sestrin2 regulates cell homeostasis and is an upstream signaling molecule for several signaling pathways. Sestrin2 leads to AMP-activated protein kinase- (AMPK-) and GTPase-activating protein activity toward Rags (GATOR) 1-mediated inhibition of mammalian target of rapamycin complex 1 (mTORC1), thereby enhancing autophagy. Sestrin2 also improves mitochondrial biogenesis via AMPK/Sirt1/peroxisome proliferator-activated receptor-gamma coactivator 1 alpha (PGC-1α) signaling pathway. Blockade of ribosomal protein … Show more

“…Both doses of FF decreased mRNA levels of Nrf2 in young but not old animals ( Figure 4 a), however, FF did not affect NRF2 protein levels in either young or old rats ( Figure 4 b). Next, we analyzed the protein levels of sestrin 2 (SESN2), which stimulates NRF2 by targeting its repressor Kelch-like ECH-Associated Protein 1 (Keap1) for autophagosomal degradation [ 11 , 12 ]. Sesn2 mRNA expression levels were similar in young and old control rats ( Figure 4 a), whereas SESN2 protein levels were two-fold higher in the old animals ( Figure 4 b).…”

“…Due to the complexity of the regulatory network, many factors and receptors can be involved in this regulation. For example, Sestrin2 activates the AMPK/Sirt1/PGC-1α pathway [ 40 ], and the downregulation of the reninogen receptor PRR can improve DKD by normalizing PGC-1α levels [ 9 ].…”
