Sesamol Inhibited Melanogenesis by Regulating Melanin-Related Signal Transduction in B16F10 Cells

Abstract: Melanin is synthesized through a series of interactions catalyzed by melanogenic enzymes such as tyrosinase, dopachrome tautomerase (tyrosinase-related protein-2; TRP-2), and tyrosinase-related protein-1 (TRP-1). Tyrosinase plays a key role in catalysing the initial and limiting steps of melanogenesis. The melanin that results from melanogenesis has the protective effect of absorbing ultraviolet radiation. However, overproduction of melanin, in addition to altering the appearance of skin, may lead to skin diso… Show more

“…Melanin overproduction was found to stimulate the cellular tyrosinase [ 35 ]. For this reason, reduction of tyrosinase activity is an efficient strategy in development of anti-melanogenic agents [ 36 , 37 ]. l -Tyrosine and l -DOPA are sequentially generated substrates that positively regulate melanogenesis and modulates melanocyte function through overlapping substrates [ 38 ].…”

“…The distribution of melanin in the skin is completed by melanosome transport to the cell membrane and transfer to neighboring keratinocytes (Beaumont et al, 2011;Wu et al, 2018). While KIF5b contributes to outward melanosome transport along microtubules (Hara et al, 2000;Hirokawa et al, 2009), Rab27A-melanophilin-myosin Va complexes regulate melanosome transport along actin filaments (Raposo and Marks, 2007).…”

“…Melanin biosynthesis occurs in melanocytes, and its storage is restricted to melanosomes, which are melanocyte-specific lysosome-related organelles (Marks and Seabra, 2001). The mature melanosomes migrate along the microtubules and actin filaments from the perinuclear area to the cell membrane and are eventually transferred to neighboring keratinocytes (Beaumont et al, 2011;Wu et al, 2018;Makbal et al, 2020).…”

“…Melanin performs a significant part in defending human skin from the damaging impacts of radiation from UV sunlight and in scavenging toxic drugs and chemicals. However, overproduction of melanin may result in skin disorders such as melasma, solar lentigo, and postinflammatory hyperpigmentation, in relation to changing the shape of the skin …”

“…However, overproduction of melanin may result in skin disorders such as melasma, solar lentigo, and postinflammatory hyperpigmentation, in relation to changing the shape of the skin. 24 Visible skin, hair, and eye pigmentation are primarily based on the different functions of melanocytes, a tiny subset of cells specializing in the synthesis and distribution of pigmented biopolymer melanin. Melanin is the main skin, hair, and eye color determinant.…”
