Serum Somatomedin/Insulin-Like Growth Factor (IGF) and IGF Carrier Levels in Patients with Cushing's Syndrome or Receiving Glucocorticoid Therapy*

Abstract: Serum levels of somatomedin (SM)/insulin-like growth factor (IGF) and IGF carrier (IGF CP) have been determined in 49 patients with Cushing's syndrome before and/or after treatment and in 11 subjects under glucocorticoid therapy.Serum samples were gel filtered in acetic acid in order to separate IGFs from their carriers. IGF content was measured by a competitive protein-binding assay using the specific IGF CP produced by rat liver in culture and human IGF ([ 125 I]IGF I as tracer and a partially purified IGF p… Show more

“…Increased 41 or unchanged 42 plasma cortisol levels have been observed after alcohol intake. IGF-I levels may be reduced 43 or unaffected 44 by corticosteroid excess. IGFBP-1 levels have been shown to decrease 45 or increase 46 during cortisol administration.…”

“…Similarly, IGFBP-3, which is GH dependent, has also been reported as low, normal or increased in glucocorticoid-treated children (3, 9±12). Thus another mechanism has been postulated, namely that glucocorticosteroids may directly inhibit osteoblastic activity in bone and cartilage, inducing a state of IGF resistance affecting bone growth (10,13,14).…”

“…19 GCs are also believed to inhibit autocrine/paracrine IGF-1 synthesis and secretion, blocking transcription by reducing IGF-1 mRNA abundance. 20,21 GC also inhibits bone growth by downregulating IGF-1, but not IGF-2, expression at the epiphyseal plate.…”
