Serum metabolic traits reveal therapeutic toxicities and responses of neoadjuvant chemoradiotherapy in patients with rectal cancer

Abstract: Neoadjuvant chemoradiotherapy (nCRT) has become the standard treatment for patients with locally advanced rectal cancer (LARC). Therapeutic efficacy of nCRT is significantly affected by treatment-induced diarrhea and hematologic toxicities. Metabolic alternations in cancer therapy are key determinants to therapeutic toxicities and responses, but exploration in large-scale clinical studies remains limited. Here, we analyze 743 serum samples from 165 LARC patients recruited in a phase III clinical study using un… Show more

“…Metabolomics is defined as the analysis of intermediate or end products of metabolic processes (metabolites) [75]. It is an easily obtained and minimally invasive method for response prediction, with serum as the preferred sample for analysis, but the literature about its role for the prediction of treatment response in patients with rectal cancer is scarce [76]. The immunoscore was originally developed [68] and later validated [69] for prognostic outcomes in colorectal patients undergoing surgery without neoadjuvant therapy.…”

“…Furthermore, these biomarkers failed to discriminate CRC from GC which led to different treatment protocols and prognosis evaluations . Metabolites, originating from the upstream products as well as containing eventual physiological information, provide a real-time understanding of disease progression. , Serum metabolites serving as a repository for all functional information on the organism can reveal the metabolic disorders of malignant tumor development and metastasis . Mass spectrometry featuring high sensitivity, simultaneous separation and identifications, and accessible databases is an advantageous tool for metabolite analysis .…”
