Serum long noncoding RNA HOTAIR as a novel diagnostic and prognostic biomarker in glioblastoma multiforme

Abstract: Glioblastoma multiforme (GBM) is the most common and aggressive malignant adult primary brain tumor. Despite surgical resection followed by radiotherapy and chemotherapy, the median survival rate is approximately 14 months. Although experimental therapies are in clinical trials for GBM, there is an urgent need for a peripheral GBM biomarker for measuring treatment response. As we have previously demonstrated that the long noncoding RNA HOX Transcript Antisense Intergenic RNA, or HOTAIR, is dysregulated in GBM … Show more

“…In recent years, the roles of lncRNAs in cancer have been widely studied. Many reports have shown that lncRNAs participate in several cancer-associated processes including chemoresistance [26,27], and abnormal expression of lncRNAs have been shown to be involved in the malignant process of glioma [28][29][30]. However, lncRNAmediated TMZ-resistant mechanisms have rarely been studied.…”

“…Genome‐wide lncRNA expression profiles in glioma patients have revealed that dysregulated lncRNA expression plays important roles in the tumorigenesis and malignant progression of this cancer, and several lncRNAs are related to the prognosis of patients with GBM 45. Circulating levels of HOTAIR are significantly correlated with high‐grade brain tumours, and one study demonstrated that this lncRNA could be considered a novel prognostic and diagnostic biomarker for GBM 97. Moreover, in vitro and in vivo studies have demonstrated that aberrant expression of lncRNAs is correlated with response to therapy in gliomas.…”

For example, knockdown of MALAT1 increases permeability of the blood-tumour barrier, which might contribute to the improvement of therapeutic strategies, 98 while restoration of CASC2 expression upregulates PTEN and can increase glioma sensitivity to temozolomidebased chemotherapy.…”

“…These discoveries suggested that lncRNAs could be excellent prognostic biomarkers and potential therapeutic targets for GBM. Although recent studies have identified some lncRNAs that exert regulatory activities during the development of GBM, these have mainly focused on the expression pattern of lncRNAs (9,10). At present, the regulatory mechanism of the vast majority of lncRNAs in GBM, especially dNA methylation, remains unclear.…”
