Serum Levels of Substance P and Response to Antidepressant Pharmacotherapy

Abstract: Substance P (SP) is possibly involved in the etiopathology of affective disorders. Here we investigated the relationship of SP serum levels and response to antidepressant drug therapy. SP serum levels were determined before and during a 9-week drug trial in 40 depressed patients treated with paroxetine in combination with either lamotrigine (n = 20) or placebo (n = 20). Responders (n = 18) and non-responders (n = 22) significantly differed in SP serum levels: responders started with higher SP levels that decre… Show more

“…47 While SSRI treatment of depression does not specifically target neuropeptide receptors, recent preliminary evidence suggests that higher pre-treatment serum substance P levels are associated with clinical response to SSRIs in depression. 48 It remains to be investigated whether SSRI treatment in asymptomatic HIV delays disease progression in association with levels of substance P. To what extent substance P is a reliable predictor of treatment response to SSRIs in HIV requires further exploration. Once again, the relationship between HIV and psychiatric illness is a bidirectional one.…”

“…Indeed, even minute elevations in circulating substance P can lead to anxiety, depression and aggression [86]. Conversely, those who respond to antidepressant pharmacotherapy are known to have declines in serum substance P in conjunction with improved mood states [87]. Thirty years have passed since it was discovered that biologically active peptides such as substance P not only communicate within the gut, brain and skin, they are also of common embryonic origin [88].…”

“…Indeed, even very slight elevations in circulating substance P can lead to anxiety, depression and aggression [81]. Conversely, those who respond to antidepressant pharmacotherapy are known to have declines in serum substance P in conjunction with improved mood states [82]. An additional area of relevance between emerging probiotic research and depression/anxiety involves the cannabinoid receptors.…”
