BackgroundOxytocin (OXT) plays a signi cant role during pregnancy, especially toward the end of pregnancy. Some studies have reported that OXT is involved in the stimulation of steroidogenesis in several organs. However, the effects of OXT on placental steroidogenesis have not yet been established. MethodsIn this study, we investigated the regulation of steroid hormones and steroidogenic enzymes by OXTassociated signaling in vitro and in vivo by western blotting analysis, ELISA and immunocytochemical analysis. ResultsOXT increased the gene expression of steroidogenic enzymes, which convert pregnenolone to progesterone and dehydroepiandrosterone (DHEA) in vitro. In OXT-administered pregnant rats, pregnenolone and DHEA levels were signi cantly enhanced in the plasma by 2.73-fold and 1.53-fold, respectively and the expression of the enzymes synthesizing DHEA, testosterone, and estradiol (E2) was increased in placental tissues. Furthermore, OXT was found to affect placental cell differentiation, which is closely related to steroid hormone synthesis. After treatment of the pregnant rats with atosiban, an antagonist of the OXT receptor, the concentration of E2 in the plasma and the expression of E2synthesizing enzyme were reduced 1.71-fold and 2.38-3.16-fold, respectively. This regulation may be due to OXT-mediated differentiation, because OXT increases the expression of corticotropin-releasing hormone, which is a biomarker of placental cell differentiation. ConclusionsOur ndings suggest that OXT regulates the differentiation of placental cells, expression of steroidogenic enzymes in the placenta, and production of critical steroid hormones during pregnancy, and it contributes to the maintenance of pregnancy.

Background The rate of cesarean section is increasing from 15% to 40% in recent years. The type of delivery influences the hormonal, emotional and mental health of the mother, which correlates positively or negatively with the health benefits of the newborn. There have been many published studies on kangaroo mother care for mothers undergoing elective cesarean section, but there is still a lack of understanding about emergency cesarean section. Objective This study aimed to evaluate the benefits of KMC practice for mothers undergoing emergency cesarean section in terms of hormone levels such as oxytocin, cortisol, prolactin, interleukin-6 and interleukin-10. Methods This retrospective analysis used remnant serum (KMC group = 16; control group = 9), and clinical data such as KMC practice (15–30 minutes three or four times daily until discharge), breastfeeding initiation time, feeding time, crying time and hospital stay were extracted from the electronic database. All hormone and cytokine expressions were quantitatively determined by ELISA. Comparisons within and between groups were performed using appropriate statistical tests. Results In the KMC group, increased levels of the hormone oxytocin significantly reduced cortisol and IL-6 and negatively influenced prolactin and IL-10 levels. The elevated prolactin facilitates overall lactation behaviour, of which 64% reported breastfeeding directly at the breast. The cytokine analysis revealed a reduction in pro-inflammatory cytokines and thereby an improved wound healing was seen in the KMC group. Conclusion These quantitative results strongly encourage the use of KMC for mothers undergoing emergency cesarean section. These simple yet effective breastfeeding strategies promote maternal and infant health, which can reduce medication use.

Background: Uterine stimulants are medications used to induce/augment labor, induce abortion, and to prevent postpartum hemorrhage. Currently, available drugs carrying adverse effects, hence the present study is designed to study the effect of Carica papaya raw fruits extract in comparison with oxytocin using in vitro rat uterus preparations. Aims and Objectives: Primary and secondary aims of the present study were to evaluate the uterine stimulant effect and also the potency of C. papaya fruit extract using in vitro rat uterus preparations in comparison with oxytocin. Materials and Methods: A total of 6 female rats were taken and anesthetized with ether. By giving midline incision at abdomen from pubic symphysis, two horns of uterus were identified and collected from each rat. Uterus of each rat was subjected to Oxytocin 0.32 IU (standard), alcoholic extract of C. papaya 0.32 mg (AECP), and combination of various doses of AECP (0.64 mg, 1.28 mg, and 2.56 mg) and a fixed dose of Oxytocin (0.32 IU). Using Sherrington’s kymograph, the uterine responses of each drug and combinations were studied in terms of height of the graph. Results: The mean contraction of uterus was 6.55mm and 2.26 cm, when Oxytocin (0.32 UI) and AECP (0.32 mg) injected, respectively. When combination of Oxytocin and different doses of AECP was injected, the uterus has shown dose-dependent response, i.e., 7.13 cm (Oxytocin 0.32 IU +AECP 3.2 mg), 8.08 cm (Oxytocin 0.32 IU + AECP 6.4 mg), and 9.01 cm (Oxytocin 0.32 IU + AECP 12.8 mg). Conclusion: AECP has shown dose-dependent uterine stimulant effect. Further studies are required to find out the possible mechanism behind the uterine stimulant activity of AECP.
