Serum concentrations of free ubiquitin and multiubiquitin chains

Abstract: Ubiquitin, which can conjugate with cellular proteins, is classified into two forms: free ubiquitin and multiubiquitin chains. The latter is active as a signal for degradation of the targeted proteins. We found both forms in human serum and, using two immunoassays, quantitated them in sera from healthy subjects and patients with some diseases. Because of putative leakage of erythrocyte ubiquitin, hemolytic serum and serum obtained after long incubation (>1–2 h) of blood at room temperature were excluded… Show more

“…The sequence of purified free Ub from secretions of stimulated chromaffin cells and Ub-derived peptide (65)(66)(67)(68)(69)(70)(71)(72)(73)(74)(75)(76), resulting from the digestion of Ub with endoproteinase Glu-C, was determined in our laboratory by automatic Edman degradation on an Applied Biosystems (Perkin Elmer, Boston, MA) 473A sequencer at the picomolar range. Samples purified by HPLC were loaded on polybrene-treated and precycled glass-fiber filters (18).…”

“…In addition, the synthetic peptide corresponding to Ub 65-76 is active against the yeast forms of C. albicans, C. tropicalis, C. neoformans, and C. glabrata with a MIC varying from 15 to 20 µM. These experiments indicate a lytic effect of Ub [65][66][67][68][69][70][71][72][73][74][75][76] against Gram-positive bacteria and fungi, because we could not obtain a restart of the microorganism growth after 48 h. Furthermore, as other antimicrobial peptides (36)(37)(38), 50 µM Ub 65-76 is unable to induce the lysis of sensitive mammalian cells like erythrocytes at a concentration up to 50 µM.…”
