Serum bile acids profiling by liquid chromatography-tandem mass spectrometry (LC-MS/MS) and its application on pediatric liver and intestinal diseases

Abstract: BackgroundA method for bile acid profiling measuring 21 primary and secondary bile acids in serum samples was developed and validated with liquid chromatography-tandem mass spectrometry (LC-MS/MS). Sample preparation included spiking with internal standards followed by protein precipitation, centrifugation, drying under nitrogen gas and reconstitution. Extracted samples were injected onto a Phenomenex Kinetex C18 column (150 × 4.60 mm, 2.6 μm).MethodsData was collected with LC-MS/MS operated in negative ion mo… Show more

“…The majority of HPLC-MS/MS methods aimed at the analysis of major five BAs (i.e., CA, CDCA, DCA, LCA, UDCA) and their glycine and taurine conjugates with analysis times under 20 min and limits of detection in the nanomolar range [30,35,47,67,80]. Scherer et al [67] developed a rapid method (runtime of 6.5 min) for quantification of 18 BAs (free and conjugated).…”

“…Due to the wide-ranging and important functions of BAs, BA pro le analysis is garnering increasing attention in medicine and zootechnics (Fu et al 2020;Lin et al 2020). However, neither the reason for nor the effect of changes in BA pro les has received su cient attention in sh.…”

“…Even a slight difference in BA hydroxylation patterns may exert drastically different biological responses (Li and Chiang 2009;Song et al, 2015). Therefore, BA pro le assessments are garnering increasing interest to screen potential biomarkers for the early diagnosis of human liver and intestinal diseases (Fu et al 2020;Slopianka et al 2017), malnutrition (Lin et al 2020), and healing processes (Roda et al 2019). It was noticed that the BA pro les of sh could be in uenced by diet (Zhang et al 2017), however, it was unclear about the agent in diet that exerted the effect.…”

“…The most abundant fragment ions were chosen for quantification purpose. Due to the lack of specific fragment ions, quantification of some unconjugated BA (UDCA, HDCA, CDCA, DCA) was accomplished by monitoring the same m/z values for both the precursor ion and the fragment ion, consistent with published literature [19]. LC conditions were optimized to achieve an optimal separation of three isobar groups of BA ( Figure 2).…”
