Serum antibody level to S-antigen in children with chronic uveitis.

Abstract: SUMMARY Bovine S-antigen was purified by gel filtration and ion exchange chromatography according to previously described techniques. An enzyme linked immunosorbent assay (ELISA) using antiserum to bovine S-antigen raised in guinea-pigs was employed to detect S-antigen in the chromatographic fractions. The purity of the S-antigen was determined by SDS-PAGE electrophoresis, where a single band was found. The purified S-antigen in microgram quantities together with Freund's complete adjuvant induced uveitis in r… Show more

“…The occurrence of such antibodies in healthy subjects may be a natural phenomenon and is probably without harm. However, sharp increases in anti-S-antigen antibody titers have been reported to occur particularly in very active forms of uveoretinitis [31,32]. We found no evidence for enhanced humoral immune responsiveness to bovine S-antigen or opsin in the patient groups suffering from serpiginous choroiditis, APMPPE or retinitis pigmentosa.…”

“…In a recent study, antibody to antigen S was demonstrated in some children with uveitis complicating JRA (23). The uveitogenic potential of antigen S in animal systems and its accompaniment by cellular or humoral immunity to the antigen support the possibility that such immunoreactivity in children with JRA and uveitis may be pathogenically important.…”

“…Animal models of experimental uveitis have employed a variety of agents, including soluble retinal antigen (S antigen) [2] and lens crystallins [3]. Studies in this laboratory [4] and others [5] have demonstrated humoral and cellmediated immunity to bovine S antigen in some children with JRA and uveitis. Studies of immunoreactivity to lens crystallins in children with JRA have not been reported.…”
