Serum and intestinal isotype antibody responses to Wa human rotavirus in gnotobiotic pigs are modulated by maternal antibodies.

Abstract: The effects of passive antibodies on protection and active immune responses to human rotavirus were studied in gnotobiotic pigs. Pigs were injected at birth with saline or sow serum of high (immunized) or low (control) antibody titre and subsets of pigs were fed colostrum and milk from immunized or control sows. Pigs were inoculated at 3-5 days of age and challenged at 21 days post-inoculation (p.i.) with virulent Wa human rotavirus. Pigs receiving immune serum with or without immune colostrum/milk were partia… Show more

“…IgM, IgG, and IgA HRV antibody geometric mean titers (GMTs) and total IgA concentrations in serum were measured by enzyme-linked immunosorbent assay (ELISA) pre-virHRV inoculation (PFMTD9 or PFMTD10/PID0), post-virHRV inoculation (PID9), and at the termination of the experiment (PID14) as described previously (16,21,74). The same parameters were measured in the small intestinal contents at PID14.…”

“…The ELISPOT assay for measuring the total immunoglobulin-secreting cell (IgSC) response and data reporting followed a previously described protocol (16). To determine the HRV-specific IgA and IgG antibody titers in serum and intestinal contents, isotypespecific ELISA was performed according to the protocol previously described (22,23). Total IgM, IgA, and IgG antibody titers in serum and intestinal contents were assessed by following an established ELISA protocol (24).…”

“…Serum from pigs that did not shed virus and serum and intestinal contents from pigs inoculated intravenous with virulent HRV were tested by antibody-ELISA as previously described for seroconversion to immunoglobulin M (IgM), IgA, and IgG antibodies to HRV (1,24,32).…”
