Serum- and Glucocorticoid-Regulated Kinase 1 Regulates Ubiquitin Ligase Neural Precursor Cell-Expressed, Developmentally Down-Regulated Protein 4-2 by Inducing Interaction with 14-3-3

Abstract: Serum- and glucocorticoid-regulated kinase 1 (SGK1) is an aldosterone-regulated early response gene product that regulates the activity of several ion transport proteins, most notably that of the epithelial sodium channel (ENaC). Recent evidence has established that SGK1 phosphorylates and inhibits Nedd4-2 (neural precursor cell-expressed, developmentally down-regulated protein 4-2), a ubiquitin ligase that decreases cell surface expression of the channel and possibly stimulates its degradation. The mechanisti… Show more

“…The effects of AII signaling in renal epithelia are further supported by the results of renal transplantation between wildtype mice and mice deficient for Agtr1a, which showed that the absence of renal AT1R reduces blood pressure (46). These findings are also consistent with studies demonstrating that phosphorylation can either negatively (47) or positively (48) regulate the activity of E3 ubiquitin ligases. KLHL2, which can also bind WNK4 (49), may be regulated by the same mechanism, because the segment spanning S433 in KLHL3 is virtually identical in KLHL2 (Fig.…”

“…Similar cooperative effects were seen on Nedd4-2-dependent ubiquitin ligase activity when using ENaC as substrate ( Fig. 3B): ENaC (␣-subunit) was heavily ubiquitinated in a Nedd4-2-dependent fashion, and SGK1 had a substantial inhibitory effect, as has been shown (24,25). We also observed modest but consistent inhibition of Nedd4-2 by GILZ1 alone (Fig.…”

“…Alanine mutations at these three sites diminish SGK1 and PKA-stimulated ENaC cell surface expression and activity (1,14). SGK1 and/or PKA-mediated Nedd4-2 phosphorylation-dependent interaction with 14-3-3 scaffolding proteins prevents subsequent interaction with and ubiquitination of ENaC (15)(16)(17). Aldosterone stimulates SGK1 expression, whereas vasopressin activates PKA.…”
