Serum and cerebral spinal fluid levels of chemokines and Th2 cytokines in Schistosoma mansoni myeloradiculopathy

Abstract: Schistosomal myeloradiculopathy (SMR) is the most common neurological form of Schistosoma mansoni infection. In this study we investigated the expression of chemokines and Th2 cytokines in serum and cerebral spinal fluid (CSF) of SMR patients. SMR patients presented increased serum levels of CCL11/eotaxin and CCL24/eotaxin-2 when compared to controls. SMR patients also had higher levels of IL-13 in CSF. Thus, SMR patients present enhancement of both IL-13 and CCR3 acting chemokines, both of which may facilitat… Show more

“…The main possible weakness was the use of a convenience sample, which makes homogeneity among the selected groups very difficult to obtain, since the levels of proteinuria, the presence and previous use of immunosuppressive drugs and the stage of disease activity at the time of collection may interfere with cytokine and chemokine measurements. Nevertheless, some aspects of the study may increase the strength of our findings, such as the utilization of strictly defined inclusion and exclusion criteria and a well-established protocol for the measurements of cytokines and chemokines with very low intra-assay variability (24).…”

“…Plasma and urinary levels of MCP-1/CCL2 and IL-8/CXCL8, plasma TGF-␤ 1 and urinary RANTES/CCL5 were measured by specific enzyme-linked immunoassay (ELISA) kits (R&D Systems, Minneapolis, MN), following the manufacturer's instructions, as described elsewhere (24). Urine chemokine levels were standardized to urine creatinine measured in the same spot urine and expressed as pg/mg cr.…”

“…For chemokine analysis, plasma samples were thawed and excess proteins, mainly albumin, were removed by acid ⁄salt precipitation (Sousa-Pereira et al 2006). Briefly, an equal volume of plasma and 1.2% trifluoroacetic acid ⁄1.35 M NaCl were mixed and left at room temperature for 10 min.…”

“…However, in the case of our patient, it is difficult to choose an explanation for the spinal cord involvement. No biopsy of the spinal cord to confirm the presence of Schistosoma mansoni eggs 30 was available and hence associated vasculitis cannot be ruled out 33 . It is worth mentioning that there was extensive involvement of the spinal cord in the case presented here (the lesion extended from T1 to T11, as documented by magnetic resonance imaging), which was followed by severe spinal cord atrophy 25 .…”
