Sertoli Cell-specific Expression of Metastasis-associated Protein 2 (MTA2) Is Required for Transcriptional Regulation of the Follicle-stimulating Hormone Receptor (FSHR) Gene during Spermatogenesis

Abstract: Background: Desensitization of FSH response by down-regulation of FSHR transcription is critical for FSH action. Results: Chromatin modifier MTA2 participates in the down-regulation of FSHR transcription. Conclusion:The FSH/Ar/MTA2 cascade may serve as an indispensable negative feedback mechanism to modulate FSH transduction events in Sertoli cells. Significance: Our findings provide new insights into mechanisms by which FSH is deregulated in male infertile patients.

“…31, 32, 33 A number of putative binding sites for transcription factors were detected in the 5' regulatory region of mammalian FSHR genes. Some of these transcription factors, such as GATA-1, 34 E2F, 34 SF-1, 35 and USF-1, 36 regulate FSHR expression by binding to its 5' regulatory region.…”

“…Western blotting was carried out as described in our previous work. 49 Protein samples were prepared in ice-cold RIPA buffer (Tris-HCl 50 mM, NaCl 150 mM, Triton X-100 1% (vol/vol), sodium deoxycholate 1% (wt/vol) and SDS 0.1% (wt/vol), pH 7.5) supplemented with complete proteinase-inhibitor cocktail tablets (Roche Diagnostic, Shanghai, China). Approximately, 20 μg of total protein was separated on SDS-PAGE and transferred to nitrocellulose membrane (Millipore, Bedford, MA, USA).…”

“…49 Briefly, after deparaffinization and rehydration, antigen retrieval was performed by incubating slides in 0.01% citrate acid in a preheated water bath (95-100°C) for 30 min and then cooling to room temperature. Sections were then exposed to 0.3% hydrogen peroxide in methanol for 10 min to destroy endogenous peroxides activity, followed by incubation with primary antibodies at 4°C overnight in a moist box.…”

“…Another member of the MTA family, MTA2, is induced by testosterone as well as by follicle-stimulating hormone (FSH) in Sertoli cells. Once induced, MTA2 also represses the transcription of the FSH receptor in FSH stimulated cells as a part of a negative feedback regulatory loop [38]. Consist with this pathway, experimental downregulation of MTA2 impairs the FSH-mediated downregulation of the FSH receptor [38].…”
