SERS-Based Aptasensor for Rapid Quantitative Detection of SARS-CoV-2

Abstract: During the COVID-19 pandemic, the development of sensitive and rapid techniques for detection of viruses have become vital. Surface-enhanced Raman scattering (SERS) is an appropriate tool for new techniques due to its high sensitivity. SERS materials modified with short-structured oligonucleotides (DNA aptamers) provide specificity for SERS biosensors. Existing SERS-based aptasensors for rapid virus detection are either inapplicable for quantitative determination or have sophisticated and expensive constructio… Show more

“…Zavyalova’s group developed a paper-based device for detection of viruses with SERS. SARS-CoV-2 virus can be measured rapidly with better selevtivity [ 264 ]. In 2017, a plasmonic filter was used by Wang’s group to filtrate, capture and identify Streptomycete spores with SERS method.…”

“…A comprehensive overview of the aptameric nanosensors developed for COVID-19 diagnosis are given in Table 1 . Vlamir et al devised an aptamer-based nanosensor containing silver nanoparticles as SERS substrate to detect SARS-CoV-2 ( Figure 1 ) [11] . It is a one-step protocol that demonstrated aggregation of silver nanoparticles after the binding of labeled aptamer with incubated virus and also showed an intense SERS signal at 587 cm -1 .…”

“…SERS spectra of adenovirus, rhinovirus, and human immunodeficiency virus (HIV) were collected previously by dropping small volumes (0.5-1 μL) of these viruses on a substrate consisting of Ag nanorod arrays [ 39 ]. A SERS-based aptasensor was developed by functionalizing colloidal AgNPs with olegonucleotides for detection of SARS-CoV-2 in water at 5.5 × 10 4 TCID50/mL level [ 40 ]. A portable handheld Raman system was used to detect influenza A virus using 10 μL of sample in water applied to Ag nanorod substrates [ 41 ].…”
