SerpinB10, a Serine Protease Inhibitor, Is Implicated in UV-Induced Cellular Response

Abstract: UV-induced DNA damage response and repair are extensively studied processes, as any malfunction in these pathways contributes to the activation of tumorigenesis. Although several proteins involved in these cellular mechanisms have been described, the entire repair cascade has remained unexplored. To identify new players in UV-induced repair, we performed a microarray screen, in which we found SerpinB10 (SPB10, Bomapin) as one of the most dramatically upregulated genes following UV irradiation. Here, we demonst… Show more

“…5 A). Notably, these gene sets have been reported to be associated with chemoresistance and malignant tumor progression [10] , [31] , [32] , [33] , [34] , [35] , [36] , [37] . Moreover, genes highly expressed in the low-risk group were significantly enriched in allograft rejection, interferon-γ response, and interferon-α response ( Fig.…”

“…We studied the potential impacts of UVB irradiation, which plays a main role in the activation of TC-NER, on the desired housekeeping genes [29,[37][38][39]. Only the B2M expression level was significantly altered by the UVB irradiation (Figure 1, upper row, second panel, NT versus 0.5 h, p = 0.0039); hence, this candidate was rejected as a reference gene.…”

“…Additionally, we also uncovered genes, which may play different roles in adaptation processes of Xiang pigs. These genes include NR2E1 for behavioral defense response 12 , SERPINB8 for maintaining the mechanical stability of skin 15 , SERPINB10 involved in the UV-induced cellular response 16 , UVSSA for response to ultra-violet (UV) radiation 30 , VEGFC responsible for regulating oxygen homeostasis 6 . The positive selection signatures identified here provided new insights into the potential adaptive mechanisms in XP.…”

“…To avoid low-quality reads, the raw paired-end reads were filtered and trimmed using the NGSQC Toolkit with default parameters 16 . High-quality reads were then aligned to the Sscrofa11.1 reference sequence using the Burrows-Wheeler Aligner (BWA) with the “bwa-mem” algorithms 72 .…”
