e Contagious bovine pleuropneumonia (CBPP) is a severe respiratory disease that is widespread in sub-Saharan Africa. It is caused by Mycoplasma mycoides subsp. mycoides, a bacterium belonging to the Mycoplasma mycoides cluster. In the absence of an efficient CBPP vaccine, improved and easy-to-use diagnostic assays for recurrent testing combined with isolation and treatment of positive animals represent an option for CBPP control in Africa. Here we describe the comprehensive screening of 17 immunogenic Mycoplasma mycoides subsp. mycoides proteins using well-characterized bovine sera for the development of a novel cocktail enzyme-linked immunosorbent assay (ELISA) for laboratory use. Two recombinant Mycoplasma immunogens, MSC_0136 and MSC_0636, were used to set up a standardized cocktail ELISA protocol. According to the results from more than 100 serum samples tested, the sensitivity and specificity of the novel cocktail ELISA were 85.6% and 96.4%, respectively, with an overall diagnostic accuracy comparable to that of the Office International des Epizooties (OIE)-prescribed serological assays. In addition, we provide a proof of principle for a field-applicable, easy-to-use commercially produced prototype lateral-flow test for rapid (<30-min) diagnosis of CBPP.

The aim of the study was to determine Mycoplasma mycoides subsp. mycoides small colony variant (MmmSC) and Mycoplasma agalactiae antibodies in ruminants from different provinces/regions of Poland. Eight hundred and ten bovine serum samples were examined for MmmSC antibodies by the use of competitive ELISA (c-ELISA) and complement fixation test (CFT). ELISA was also used for M. agalactiae antibody detection in 951 serum samples of sheep and goats. The first screening serological examination of MmmSC antibodies using c-ELISA revealed two (0.25%) positive and 135 (16.92%) doubtful results. The second examination revealed only 52 doubtful results, whereas the rest samples were negative. To compare, the final confirmatory examination by CFT gave 100% of seronegative results. The examination performed in small ruminants demonstrated only one doubtful result, which was finally defined as negative following the second ELISA, whereas the remaining samples were negative. To conclude, the present serological study showed the lack of infections in Polish domestic ruminant caused by two mycoplasmas.

The lipoprotein LppQ is the most prominent antigen of Mycoplasma mycoides subsp. mycoides small colony type (SC) during infection of cattle. This pathogen causes contagious bovine pleuropneumonia (CBPP), a devastating disease of considerable socio-economic importance in many countries worldwide. The dominant antigenicity and high specificity for M. mycoides subsp. mycoides SC of lipoprotein LppQ have been exploited for serological diagnosis and for epidemiological investigations of CBPP. Scanning electron microscopy and immunogold labelling were used to provide ultrastructural evidence that LppQ is located to the cell membrane at the outer surface of M. mycoides subsp. mycoides SC. The selectivity and specificity of this method were demonstrated through discriminating localization of extracellular (i.e., in the zone of contact with host cells) vs. integral membrane domains of LppQ. Thus, our findings support the suggestion that the accessible N-terminal domain of LppQ is surface exposed and such surface localization may be implicated in the pathogenesis of CBPP.
