Skin cancer is one of the most common malignancies in white populations. The therapy strategy is important in skin cancer treatment, depending on several criteria such as stage, size, and localization. Removal of cancerous tissue following anticancer therapeutic administration is considered as gold standard in skin cancer treatment. However, annually rising drug resistance, local inflammation, and ineffective treatment result in a reduction in the effectiveness of the patient's treatment. Nanotechnology has emerged as a prospective in the field of skin cancer medicine, offering innovative, promising solutions for therapeutic procedures and targeted drug delivery. Different nanomaterials are investigated for their potential in skin cancer treatment. Nanohydrogels as a hybrid material, have gained considerable attention due to their unique biomedical and pharmaceutical properties, such as biocompatibility, high water content, and tunable physicochemical characteristics. The principal problem with common skin melanoma chemotherapy is the strong side effects because therapeutics used for treatment do not distinguish cancer cells from healthy cells. Nanohydrogels, as a new-generation, versatile system with the possession of dual characteristics of hydrogels and nanoparticles have shown great potential in targeted delivery in cancer therapy thanks to the possibility of their various modifications, and by that overcome problems with side effects of treatment. This scientific review provides an analysis of the current state of research on nanohydrogels in skin cancer medicine, highlighting their design principles, synthesis methods, and applications in drug delivery, imaging, and combination therapies.

Cancer-associated fibroblasts (CAF) are the most abundant stromal cells in the tumor microenvironment (TME), especially in solid tumors. It has been confirmed that it can not only interact with tumor cells to promote cancer progression and metastasis, but also affect the infiltration and function of immune cells to induce chemotherapy and immunotherapy resistance. So, targeting CAF has been considered an important method in cancer treatment. The rapid development of nanotechnology provides a good perspective to improve the efficiency of targeting CAF. At present, more and more researches have focused on the application of nanoparticles (NPs) in targeting CAF. These studies explored the effects of different types of NPs on CAF and the multifunctional nanomedicines that can eliminate CAF are able to enhance the EPR effect which facilitate the anti-tumor effect of themselves. There also exist amounts of studies focusing on using NPs to inhibit the activation and function of CAF to improve the therapeutic efficacy. The application of NPs targeting CAF needs to be based on an understanding of CAF biology. Therefore, in this review, we first summarized the latest progress of CAF biology, then discussed the types of CAF-targeting NPs and the main strategies in the current. The aim is to elucidate the application of NPs in targeting CAF and provide new insights for engineering nanomedicine to enhance immune response in cancer treatment.

In recent years, the incidence and mortality of breast cancer have shown a rapidly increasing trend worldwide. Gemcitabine hydrochloride (GEM) is clinically the first-line chemotherapeutic drug for breast cancer with excellent antitumor effects. However, the short half-life of GEM and the lack of sustained release effects predispose it to myelosuppression and nephrotoxicity. Red blood cells (RBCs) are primarily used as biological carriers for drug delivery. In view of the shortcomings of GEM in clinical practice, this study constructed a folic acid/gold nanoparticle (AuNPs) surface co-modified RBC carrying the GEM delivery system (FA/Au-GEM-RBCs), based on RBCs as a carrier, aiming to enhance the targeting effect of the carrier. AuNPs could be used as glucose-like oxidase to catalyze glucose oxidation into H 2 O 2 , cutting off the source of nutrients to cancer cells, while producing H 2 O 2 damages the RBCs membrane (RBCM) to release drugs. Here, AuNPs were used as a "gated switch" to promote drug release. This DDS has in vitro properties similar to natural RBCs, with the drug loading amount of 10.90 mg/10 9 RBCs and can circulate for up to 9 days in vivo. In antitumor study studies, FA/Au-GEM-RBCs could target tumor tissues with a tumor inhibition rate of 71.41% ± 8.15%. The delivery system generated multiple reactive oxygen species (ROS), significantly enhancing the effect of tumor treatment. In conclusion, the constructed FA/Au-GEM-RBCs could substantially improve the tumor-killing effect of GEM and deplete the nutrient source at the tumor site. Therefore, this system is expected to achieve synergistic treatment with chemotherapy and starvation therapy and further provide ideas for the treatment of breast cancer.
