Sequential multiplex PCR assay for determining capsular serotypes of colonizing S. pneumoniae

Abstract: BackgroundAsymptomatic nasopharyngeal carriage represents an important biological marker for monitoring pneumococcal serotype distribution and evaluating vaccine effects. Serotype determination by conventional method (Quellung reaction) is technically and financially challenging. On the contrary, PCR-based serotyping represents a simple, economic and promising alternative method.MethodWe designed a novel multiplex PCR assay for specific detection of the 30 classical colonizing S. pneumoniae serogroups/types. “…The CDC serotyping schemes have been modified in different ways for use not only in Finland but in several other countries as well, which indicates that the oligonucleotide primers used for amplification can be combined in several ways (10,15,23,24). This flexibility is important, as it allows the modification of a scheme according to the serotype distribution of the material to be serotyped, for example, that originating from different geographical regions of the world or representing colonizing or invasive isolates.…”

“…Also, the oligonucleotide primers described by Jourdain et al, and Brito et al, were used in this study. The sequences of primers [13,14] for amplification of serotypes of S. pneumonia are given in [Table/ Fig-1]. …”

“…PCR-based serotyping targeting the cps locus has been shown to be effective for the serotyping of pneumococcal specimens [3][4][5][6]. Based upon cumulative results from simultaneous serology and PCR-based testing of diverse isolate sets, a sequential PCR assay is considered accurate for the serotyping of S. pneumonia isolates [2,7].…”
