In the last 20 years, bone regenerative research has experienced exponential growth thanks to the discovery of new nanomaterials and improved manufacturing technologies that have emerged in the biomedical field. This revolution demands standardization of methods employed for biomaterials characterization in order to achieve comparable, interoperable, and reproducible results. The exploited methods for characterization span from biophysics and biochemical techniques, including microscopy and spectroscopy, functional assays for biological properties, and molecular profiling. This review aims to provide scholars with a rapid handbook collecting multidisciplinary methods for bone substitute R&D and validation, getting sources from an up-to-date and comprehensive examination of the scientific landscape.

An ideal craniofacial bone repair graft shall not only focus on the repair ability but also the regeneration of natural architecture with occlusal loads‐related function restoration. However, such functional bone tissue engineering scaffold has rarely been reported. Herein, a hierarchical 3D graft is proposed for rebuilding craniofacial bone with both natural structure and healthy biofunction reconstruction. Inspired by the bone healing process, an organic–inorganic nanoink with ultrasmall calcium phosphate oligomers and bone morphogenetic protein‐2 incorporated is developed for spatiotemporal guidance of new bone. Based on such homogeneous nanoink, a biomimetic graft, including a cortical layer containing Haversian system, and a cancellous layer featured with triply periodic minimum surface macrostructures, is fabricated via projection‐based 3D printing method, and the layers are loaded with distinct concentrations of bioactive factors for regenerating new bone with gradient density. The graft exhibits excellent osteogenic and angiogenic potential in vitro, and accelerates revascularization and reconstructs neo‐bone with original morphology in vivo. Benefiting from such natural architecture, loading force is widely transferred with reduced stress concentration around the inserted dental implant. Taken from native physiochemical and structural cues, this wstudy provides a novel strategy for functional tissue engineering through designing function‐oriented biomaterials.

Native bone inherently requires a balanced ionic microenvironment to maintain bone homeostasis. Hence, scaffolds designed for the sustained release of therapeutic ions into bone defects hold great promise for bone regeneration. Magnesium (Mg) and silicon (Si) are essential elements, which play crucial roles in the process of bone regeneration, impacting immunomodulation, angiogenesis, and osteogenesis. Herein, porous cryogel‐type organic–inorganic composite microspheres are developed as injectable microscaffolds (denoted as GMN). GMN enables sustained release of Mg/Si ions at an optimized ratio, achieving the most significant synergistic effect on vascularized bone regeneration. Various conditioned media are obtained to explore angiogenesis‐osteogenesis coupling, as well as the crosstalk between bone marrow mesenchymal stromal cells (BMSCs) and macrophages. Meanwhile, autocrine and paracrine effects simultaneously play synergistic modulating functions in determining cell fates under the guidance of Mg/Si ions and biofactors secreted by cells. Overall, the Mg/Si ion‐engineering microscaffolds create a conducive microenvironment to efficiently augment the regeneration of vascularized bone tissue in vivo, offering a versatile platform for tissue engineering.
