Sequential addition of neuronal stem cell temporal cohorts generates a feed-forward circuit in the Drosophila larval nerve cord

Abstract: How circuits self-assemble starting from neuronal stem cells is a fundamental question in developmental neurobiology. Here, we addressed how neurons from different stem cell lineages wire with each other to form a specific circuit motif. In Drosophila larvae, we combined developmental genetics (Twin spot MARCM, Multi-color Flip Out, permanent labeling) with circuit analysis (calcium imaging, connectomics, network science). For many lineages, neuronal progeny are organized into subunits called temporal cohorts.… Show more

“…Brain input neurons (Fig. 1F) comprise two broad classes: 1) sensory neurons (SNs) with axons in the brain; many were reconstructed and characterized previously (45)(46)(47)61), and 2) ascending neurons (ANs) that transmit somatosensory signals from the VNC; several were previously reconstructed (38,41,44,(62)(63)(64). We reconstructed an additional set of A1 ANs, resulting in the most complete set of ANs from a nerve cord segment (A1) to date (Fig.…”

“…To confirm the identities of unpublished neurons, we compared their morphology to light level examples ( Merritt and Whitington, 1995 ; Schrader and Merritt, 2000 ; Grueber et al, 2002 , 2007 ). For details of matching neurons in light-level data, see both Wreden et al (2017) and Wang et al (2022) . In addition, we compared each neuron in other segments to the A1 example of the same name and to its left-right hemisegment sister neuron.…”

“…Our findings reveal that multiple lineages participate in building a complex circuit. Drosophila larval ventral nerve cord cohorts were shown to be formed by sequential addition from different lineages over time, where circuit input neurons are born prior to the output neurons 97 .…”
