Sequencing and functional validation of the JGI Brachypodium distachyon T‐DNA collection

Abstract: Due to a large and growing collection of genomic and experimental resources, Brachypodium distachyon has emerged as a powerful experimental model for the grasses. To add to these resources we sequenced 21 165 T-DNA lines, 15 569 of which were produced in this study. This increased the number of unique insertion sites in the T-DNA collection by 21 078, bringing the overall total to 26 112. Thirty-seven per cent (9754) of these insertion sites are within genes (including untranslated regions and introns) and 28%… Show more

“…To study BdPYL1 involvement in ABA perception, we analyzed a BdPYL1 T-DNA insertion line JJ20227-Bdpyl1-1 ( Figure S7; Bragg et al, 2012;Hsia et al, 2017). T-DNA insertion in BdPYL1 includes a 4 9 35S enhancer that drives the majority of the BdPYL1 coding sequence in an anti-sense conformation.…”

“…Agrobacterium tumefaciens AGL1, a supervirulent strain, has been used for the majority of B. distachyon transformations (Alves et al., ; Bragg et al., ; Hsia et al., ; Vogel & Hill, ). However, a recent study found that A. rhizogenes strain 18r12v can also transform B. distachyon with efficiency comparable to or slightly higher than that of AGL1 (Collier et al., ).…”

“…The protocol described here has been successfully employed to create over 22,000 T‐DNA mutants (Bragg et al., ; Hsia et al., ) and has been used for studying individual genes, including the functional validation of the flowering‐time gene FT (Qin et al., ; Wu et al., ) and wheat male sterile gene Ms2 (Ni et al., ), among others.…”

“…Thus, research on model grasses such as Setaria viridis and Brachypodium distachyon can inform growth strategies for many crops (Brutnell et al, 2015). B. distachyon is gaining popularity as a model grass because of its small genome, short generation time, genetic tractability, and the availability of extensive germplasm and mutant collections (Hsia et al, 2017). Additionally, since it uses C 3 carbon (C) fixation, it is a good laboratory model plant relevant to cereal crops such as barley (Hordeum vulgare), rice (Oryza sativa), and wheat (Triticum aestivum).…”
