Sequence of Subterranean Clover Stunt Virus DNA: Affinities with the Geminiviruses

Abstract: The nucleotide sequences of seven circular, single-stranded DNA components of one isolate of subterranean clover stunt virus (SCSV) have been determined. Each component, of about 1 kb, appears to encode a single open reading frame in the same sense as the encapsidated DNA. Notably, the proteins encoded by two SCSV components are related. Each has a consensus nucleotide binding motif and shares about 40% amino acid identity with the other and with the putative replication proteins of banana bunchy top virus and… Show more

“…Cloning of genomic components has revealed the genomic components of the genus Nanovirus are smaller than their BBTV counterpart by approximately 100 bp (Vetten et al, 2005). Sequence analysis of the genomic components has also revealed differences between the nanoviruses and BBTV within the intergenic region, most notably the iteron sequences [Rep binding motifs and the nanonucleotide loop sequence (5' (T/C) AGTATTAC 3'; conserved in the genus Nanovirus] (Boevink et al, 1995).…”

“…BBTV shares a number of characteristics with four other plant viruses [subterranean clover stunt virus (SCSV) (Boevink et al, 1995), coconut foliar decay virus (CFDV) (Rohde et al, 1990), faba bean necrotic yellows virus (FBNYV) (Katul et al, 1995) and milk vetch dwarf virus (MDV) (Sano et al, 1993)] which all have isometric virions and circular ssDNA genomes with components of approximately 1 kb.…”

“…SCSV has a functionally divided genome composed of multiple circular, ssDNA molecules of about 1 kb, each of which are encapsidated in a separate particle with a short complementary primer sequence [13,14]. The sequence of eight circular, ssDNA components of SCSV has been determined so far [8,94]. All components contain a single ORF in sense direction and a stable stem-loop structure in the noncoding region.…”
