Sequence and expression pattern of the Stra7 (Gbx‐2) homeobox‐containing gene induced by retinoic acid in P19 embryonal carcinoma cells

Abstract: The cDNA sequence of Stra7, a retinoic acid (RA)-inducible gene in P19 embryonal carcinoma (EC) cells, was determined. The deduced Stra7 protein contains a homeodomain highly similar to that of the previously described chicken CHox7 gene product, and is highly conserved during evolution, from hemichordates to vertebrates. The mouse Stru7 cDNA corresponds to the full-length form of the 77 bp homeodomainencoding cDNA fragment which was previously cloned and termed MMoxA or Gbx-2. Reversetranscriptase-PCR analysi… Show more

“…These distributions become more symmetrical at 24 and 48 h and may represent a second wave of gene regulation mediated by the early induced genes. Several of the genes have been previously shown to be direct targets of RARs, in EC and other cell contexts and include LeftyA, CYP26A1, CRABP2, Meis2, HoxD1, midkine, Gbx2 and neurogranin (see Table 1) (OuladAbdelghani et al, 1997(OuladAbdelghani et al, , 1998White et al, 1997;Durand et al, 1992;Simeone et al, 1990;Michikawa et al, 1993;Bouillet et al, 1995a;Iniguez et al, 1994). This supports the validity in using cDNA microarray analysis to identify RA targets in EC cells.…”

“…To determine if more posterior neurectoderm forms, we examined Gbx2 expression, a marker of anterior hindbrain at 8.5 dpc (Fig. 5E) (Bouillet et al, 1995;Wassarman et al, 1997). In Lrp5 +/-;Lrp6 -/-embryos there is a stripe of Gbx2 expression in the head region, indicating the formation, but lack of expansion, of anterior hindbrain (Fig.…”
