Sequence and expression of the murine Hoxd-3 homeobox gene.

Abstract: Murine Hoxd-3 (Hox 4.1) genomic DNA and cDNA and Hoxa-3 (Hox 1.5) cDNA were cloned and sequenced. The homeodomains of Hoxd-3 and Hoxa-3 and regions before and after the homeodomain are highly conserved. Both Hoxd-3 and Hoxa-3 proteins have a proline-rich region that contains consensus amino acid sequences for binding to Src homology 3 domains of some signal transduction proteins. Northern blot analysis of RNA from 8-to 11-day-old mouse embryos revealed a 4.3-kb species of Hoxd-3 RNA, whereas a less abundant 3.… Show more

“…The genes contain a homeobox, a 183-bp sequence encoding a 61-amino acid sequence that has a DNA-binding motif for transcriptional regulation of sofar nondefined genes. Some of the homologs of Hox genes that exist in Drosophila are expressed in the meso-and metane-phros, such as Hoxd-3 (263). Although Hox genes are believed to be involved primarily in regional patterning during embryogenesis, their specific roles in kidney organogenesis remain to be discovered, since very few of the targeted mutations so far have resulted in renal defects (149), suggesting redundant gene functions in the cluster or nested expression domains.…”

“…S2a,b). These results indicate that, in addition to endogenous Hoxd3 expression from the caudal myelencephalon to the spinal cord (Tan et al. 1996), the HOXD3 gene exogenously introduced is excessively expressed in the neural tube of the transgenic embryos.…”

“…Second, because the amino acid sequence of the HOXD3 protein is 95% identical with that of the mouse Hoxd3 protein (Taniguchi et al. 1992; Tan et al. 1996), it was expected that there would be little difference in function between the two proteins.…”

“…Other Hox genes, such as Hoxa-5, Hoxa-6, Hoxd-3, and Hoxd-4, are expressed in the mye!encephalon (Dony and Gruss, 1987;Toth et al, 1987;Gaunt et al, 1989;Tan et al, 1996). Hoxa-4 likewise is detected in the mye!encephalon except that at El5.5 the boundary of expression extends anteriorly next to the limit between medulla and pons (see Fig.…”
