Sequence analysis of the ribosomal DNA ITS2 region in two Trichogramma species (Hymenoptera: Trichogrammatidae)

Abstract: Two egg parasitoid wasps, Trichogramma euproctidis (Girault) and Trichogramma brassicae (Bezdenko) (Hymenoptera: Trichogrammatidae) were identified in the study. The taxonomy of these wasps is problematic because of their small size and lack of distinguishable morphological characters. The DNA sequence variation from the internal transcribed spacer 2 (ITS2) region of nuclear ribosomal DNA (rDNA) was analyzed from these two Trichogramma species. This technique provides quick, simple and reliable molecular… Show more

“…The ITS region of the highly repeated nrDNA gene family has widely proven useful and powerful in distinguishing among closely related species and even between cryptic species of Trichogramma (Ercan et al, 2011;Poorjavad et al, 2012). This is mostly due to its generally similar sequence pattern within species (Vogler & DeSalle, 1994).…”

“…We first analyzed the phylogenetic backgrounds of the two strains. ITS-2 gene has widely proven to be a powerful tool in distinguishing among closely related Trichogramma species 42,6366 . Its usefulness for species identification in Trichogramma is because: (1) smaller divergence within species than between species; (2) morphologically distinct cryptic species can be discriminated by sequence differences 42 .…”

“…rDNA genes or nucleotide regions have proven to be useful in taxonomic studies for various taxa and one of the most important target regions used in molecular analysis was ribosomal DNA which is relatively less changed and is composed of 28S D2 gene regions and internal spaces ITS2. The sequence analysis of ITS2 rDNA has been described in several studies as a tool for Trichogrammatidae identification as well (Kan et al 1996;Pinto et al 1997Pinto et al , 2002Sayed et al 2011;Sumer et al 2011). 28S rDNA coding rRNA is accepted as a highly conserved gene region during evolutionary processes, so it is used to construct phylogenetic trees especially for higher categories.…”
