Sequence analysis of cDNA coding for a major house dust mite allergen, Der f I

Abstract: A cDNA clone coding for Der f I, a major allergen from the house dust mite Dermatophagoides farinae has been isolated and sequenced. It codes for a putative 18-residue signal peptide, an 80-residue proenzyme region, and a 223-residue mature protein with a derived molecular weight of 25,191. The deduced amino-acid sequence shows significant homology to other cysteine proteases in the proregion as well as in the mature protein. Sequence alignment of the mature Der f I protein with the homologous allergen Der p I… Show more

“…Group 1 and group 2 allergens are major allergens derived from house dust mites based on the frequency of patients sensitized, amount of specific IgE, and content in mite extract (4 -10). The group 1 allergens, Der p 1 from D. pteronyssinus and Der f1 from D. farinae, belong to the papain-like cysteine protease family (11,12) and actually exhibit cysteine protease activity (13)(14)(15)(16). The cDNAs for Der p 1 and Der f 1 were isolated when cDNA cloning of allergens began (11,12,17).…”

“…A few examples of wellknown allergens characterized in this way are ragweed allergens Amb a 1 [1] and Amb a 2 [2], grass pollen allergens Lol p 1 [3,4] and Lol p 2 [5], house dust mite allergens Der p 1 [6] and Der f 1 [7], peanut allergen Ara h 1 [8], shrimp allergen Met e 1 [9], and latex allergen Hev b 1 [10]. This technology has the potential to be useful in several ways.…”

“…As complex organisms, house dust mites produce thousands of different proteins and macromolecules, and over 30 IgE-binding components have been demonstrated in patients allergic to house dust mites by immunological analysis (2). Of these allergens, group 1 is the major one and has been identified to be homologous with cysteine proteases (3).…”
