Separation of Native Allophycocyanin and R-Phycocyanin from Marine Red Macroalga Polysiphonia urceolata by the Polyacrylamide Gel Electrophoresis Performed in Novel Buffer Systems

Abstract: Three buffer systems of Imidazole−Acetic acid, HEPES−Imidazole/Bis-tris and Bis-tris−HEPES−MES were designed based on the principle of discontinuous polyacrylamide gel electrophoresis (PAGE) for the native PAGE which could be performed in pH 7.0 and 6.5 in order to analyze and prepare the minor components of allophycocyanin (AP) and R-phycocyanin (R-PC) from marine red macroalga Polysiphonia urceolata. These AP and R-PC phycobiliproteins are easily denatured in alkaline environments. The obtained results demon… Show more

“…However, PC-β was not observed in the protein samples analyzed in the present study. PC in seaweed is usually a monomer with a molecular weight of 40 kDa, and is composed of an α-chain with a molecular weight of 18.5 kDa and β-chain with a molecular weight of 20.5 kDa, and two or three blue-colored phycocyanobilin molecules bind with one phycoerythrobilin molecule ( 35 , 36 ). PC in red algae is the R-type with an absorption band between 615 and 620 nm, however, certain types show an absorption band at 550 nm, caused by phycoerythrobilin ( 33 ).…”

“…In APC, a monomer with a molecular weight of 15.5 kDa forms a hexamer ( 35 , 36 ). APC is less abundant than PE or PC and, like PC, has a bond between phycocyanobilin and the protein.…”

“…Further, these concentrations were compared with past investigations of indoor dust levels in households, schools, and offices, along with soil, surface, and street dust in rural and urban areas across the globe (Table 3). From the Table 3, it is clear that Fe, Mn, Co, Cr, Ni, and Pb levels were comparable to household dust (Chattopadhyay, Lin, and Feitz 2003;Kelepertzis et al 2019;Kurt-Karakus 2012;Yoshinaga et al 2014), school dust (Figueroa, De la O-Villanueva, and De la Parra 2007), and street dust (Du et al 2013;Ferreira-Baptista and De Miguel 2005;Masto et al 2017;Pathak et al 2013;Yu, Wang, and Zhou 2014). Zn levels were significantly lower and similar to surface soils (Li et al 2016) and household dust (Madany et al 1994).…”

“…The peak at approximately 550 nm comes from phycoerythrobilins and conspicuously expressed absorption maximum at approximately 615-620 nm is related to phycocyanobilins. Allophycocyanin has an absorption maximum at 650 nm [5][6][7][8]. R-PC from the marine red macroalga Polysiphonia urceolata (Lightfoot ex Dillwyn) Greville is composed of one α and two β subunits.…”
