The distribution of intraperitoneally administered [4-14C]pregnenolone and [1 ,2-3H]corticosterone were studied in male and female conventional rats at different times after injection of the isotopes. The administered steroids were rapidly excreted into the intestinal tract ; 2 h after administration 60-700/, of the given dose was localized in the small bowel, mainly in monosulphurylated form.The elimination in urine and faeces of intraperitoneally administered [4-14C]pregnenolone and [4-14C]corticosterone was studied in germfree and conventional, male and female rats. The steroids were excreted as free and conjugated metabolites. The latter were mainly monosulphurylated, and to a smaller extent disulphurylated. No steroid glucuronides were found. As a probable result of the intestinal microbial sulphohydrolase activities conventional rats excreted more free steroids than germfree both in faeces and urine. Male rats excreted more unconjugated and more polar steroids than female. Corticosterone metabolites were monosulphurylated to a larger extent than pregnenolone metabolites ; faecal excretion of corticosterone metabolites was quantitatively more important than of pregnenolone metabolites.I n a series of publications we have described the great differences in the steroid hormone metabolite pattern in faeces from germfree and conventional rats (for references, see [I] -pregnene-3,11,15,20-tetrone were kindly given by Dr. J. Babcock (Upjohn Co., Michigan, U.S.A.). The purity of these reference steroids was analyzed by thin-layer and gas-liquid chromatography. MATERIALS AND METHODS ReferenceDigestive juice from Helix pomatia was purchased from Industrie Biologique FranOaise (Gennevilliers, France). Animals and Collection of Paeces and UrineFive male and five female germfree rats were kept in metabolism cages. Five male and five female control rats of the same stock were reared in metabolism cages outside the germ free isolators on the same sterilized diet as the germfree animals. These rats were used in two experiments. Maternal effects have gained attention as a method by which mothers may alter the physiological condition and phenotype of their offspring based upon current environmental conditions. The physiological and phenotypic outcomes of glucocorticoid-mediated maternal effects have been extensively studied in a variety of vertebrates; however, the underlying mechanism is currently unclear. Here, we injected tritiated corticosterone into the yolks of freshly laid Japanese quail eggs (Coturnix japonica) and traced its movement and metabolism through the in ovo development period. We found that corticosterone was extensively conjugated throughout the egg by the end of development, and while minimal corticosterone was detected within the embryo during development, accumulation of a conjugated metabolite in the embryo started to occur on day 6 of development. Because no movement and metabolism of corticosterone occurred in infertile eggs, our findings suggest that embryos are not passive recipients of maternal steroids, but instead appear to possess extensive metabolic capabilities, which may modulate their exposure to maternal steroids.

The steroids in faeces from three healthy persons, one woman and two men, were analyzed by gas-liquid chromatography and gas chromatography-mass spectrometry. Both C,,O,, C,,O,, CZlO, and C,,O, steroids were found in the free steroid and in the steroid monosulphate fractions.Incubation of various precursor steroids with intestinal contents recovered from the ileostoma of a colectomized patient demonstrated the presence of intestinal bacterial 16a-and 21-dehydroxylating enzymes. Furthermore, it was shown that a A16-C,, steroid was converted to a i7a-pregnane derivative and that the side chain of lI~,21-dihydroxy-4-pregnene-3,2O-dione could be removed by the intestinal bacteria.Little is known about the excretion of steroid hormone metabolites in faeces from adult humans. New purification methods in combination with gas chromatography-mass spectrometry has made it possible to study also this pathway of steroid excretion. Recently we described the pattern of C,, and C,, steroids in faeces from pregnant women [l]. The present investigation describes the excretion of steroid hormone metabolites in faeces from two healthy men and one woman and also demonstrates, by in vitro techniques, some steroid metabolizing enzymes of the human intestinal microorganisms.
