The steroids in faeces from three healthy persons, one woman and two men, were analyzed by gas-liquid chromatography and gas chromatography-mass spectrometry. Both C,,O,, C,,O,, CZlO, and C,,O, steroids were found in the free steroid and in the steroid monosulphate fractions.Incubation of various precursor steroids with intestinal contents recovered from the ileostoma of a colectomized patient demonstrated the presence of intestinal bacterial 16a-and 21-dehydroxylating enzymes. Furthermore, it was shown that a A16-C,, steroid was converted to a i7a-pregnane derivative and that the side chain of lI~,21-dihydroxy-4-pregnene-3,2O-dione could be removed by the intestinal bacteria.Little is known about the excretion of steroid hormone metabolites in faeces from adult humans. New purification methods in combination with gas chromatography-mass spectrometry has made it possible to study also this pathway of steroid excretion. Recently we described the pattern of C,, and C,, steroids in faeces from pregnant women [l]. The present investigation describes the excretion of steroid hormone metabolites in faeces from two healthy men and one woman and also demonstrates, by in vitro techniques, some steroid metabolizing enzymes of the human intestinal microorganisms.
MATERIALS AND METHODSSteroids 5a-Pregnane-3p,20a-diol, 5-pregnene-3p,20~x-diol, 3/3-hydroxy-5-androsten-i7-one, 3a-hydroxy-5/3,17p-pregnan-20-one and 5-androstene-3p,17/?-diol were purchased from Ikapharm (Ramat-Gan, P. 0.
