Séparation des particules ribonucléoprotéiques (Hn‐RNP) des noyaux de cellules L, en deux classes, par centrifugation isopycnique en gradients de métrizamide

Abstract: SummarySeparation of nuclear Hn-RNP particles into two density classes, following isopycnic centrifugation in metrizamide gradients, is reported. One class (density 1.31 g/ml) is rapidly labelled, and contains polydisperse heterogeneous high molecular weight particles, easily diffusible from intact nuclei under certain conditions. The other class (density 1.18 g/ml) is of lower molecular weight, not diffusible, and needs apparently a longer time to be labelled and/or to accumulate inside the nuclei.

“…2-(3 acetamido-5-N-methylacetamido-2,4,6-triiodobenzamido)-2-deoxy-D-glucose (Metrizamide, Nyegaard and Co A/S, Oslo) was dissolved in 20 mM Triethanolamine-HCl, pH 7.4, 25 mM KCl, 3 mM MgCl2, 10 mM mercaptoethanol. Two methods were used for making the gradients: Method I: Fractions of sucrose gradients were pooled and 1.8 ml were layered on 3.4 ml of a solution of 40 % (w/v) of metrizamide (7).…”

“…6). In other systems messenger RNP (mRNP) usually bands in metrizamide at a lower density than the ribosomes (3,8), whereas purified RNA bands at 1.17 g/cm3. The non-adenylated RNA which sediments between 20S and 80S (Fig.…”
