Separation and determination of carnosine-related peptides using capillary electrophoresis with laser-induced fluorescence detection

Abstract: A capillary electrophoresis (CE) method with laser-induced fluorescence (LIF) detection was developed for the separation and detection of carnosine-related peptides (carnosine, anserine, and homocarnosine). A sensitive and fluorogenic regent, 3-(4-carboxybenzoyl) quinoline-2-carboxaldehyde (CBQCA) was selected as a precapillary labeling reagent for imidazole dipeptides to form isoindole derivatives. The optimized molar ratio between CBQCA and peptide was found to be 75:1, and 50 mmol/L borate buffer (pH 9.2) w… Show more

“…PepT2 may also transport homocarnosine (c-aminobutyryl-L-histidine) and although carnosine is absent from human CSF, homocarnosine is present; furthermore, CSF levels of homocarnosine have been reported to decline with age in humans by a factor of 7 or more (Huang et al 2005;Jansen et al 2006). However, the significance of these observations is uncertain.…”

“…It is interesting to note that treatment of humans with anticonvulsants gabapentin and topiramate raise levels of homocarnosine, a carnosine homologue, in cerebrospinal fluid (CSF) (Petroff et al, 1998 and2006), and that, while carnosine is absent from human CSF, the levels of CSF homocarnosine decrease markedly with age (Jansen et al, 2006;Huang et al, 2005).…”

“…However, as peptides and AAs mostly show only little or moderate UV absorbance within the wavelength range commonly used for detection, derivatization procedures are required to facilitate their sensitive detection [15]. LIF [16][17][18][19][20] detection and electrochemical detection [21,22] were another two detection techniques that had been commonly used since they possessed higher detection sensitivity than UV detector. However, derivatization procedures are usually required for LIF detection, and electrochemical detector was only used to detect electroactive substances.…”
