Separation and Characterization of Intestinal Brush Border Enzymes in Adult Rats and in Suckling Rats under Normal Conditions and after Hydrocortisone Injections

Abstract: Adult and suckling (13 days old) rat intestinal brush borders have been purified by the procedure of Schmitz et al. (25). Enzymatic proteins have been separated by polyacrylamide gel electrophoresis. In the adult rat, enzyme activities in order from the origin were: maltase/ glucoamylase/sucrase-isomaltase (protein band 3), lactase (protein band 5), maltase/sucraseisomaltase (protein band 6) and alkaline phosphatase (protein bands 8 and 9). In the suckling rat, protein band 5 associated with lactase activity w… Show more

“…The proximal part of the small intestine was resected, everted and the mucosa was removed. Partial purification of brush border membranes has been performed by a modification (14) of the procedure of Schmitz et al (18). The brush border proteins were separated by gel electrophoresis on 7.5% polyacrylamide gels containing 0.1% sodium dodecyl sulfate (8).…”

“…Enzymes were eluted from the slices by overnight incubation of each slice in 1 ml of distilled water at 4 °C. Disaccharidases [isomaltase (substrate:palatinose), lactase, maltase, sucrase) as well as glucoamylase were determined according to Messer and Dahlqvist (12) on aliquots of the gel eluates (14) and of the starting homogenates (lactase, sucrase). The enzyme activities of the ho mogenates were expressed as mU/mg protein (mean t SEM).…”

“…It is generally assumed that it is the glucocorticoid hormone which triggers the maturation. The serum levels of glucocorticoid hormones increase rapidly at the time of weaning (2) and treatment with large doses of glucocorticoid in suckling rats results in precocious development of enzymes in the gastrointestinal tract (3)(4)(5)(6).…”
