Sensitivity of various human lymphoblastoid cells to the antiviral and anticellular activity of human leukocyte interferon

Abstract: Of eight lymphoblastoid cell lines studied five were insensitive to both the anticellular and antiviral activities of human leukocyte interferon, and two were sensitive to both activities. One line could not be fully evaluated since it was not possible to study its sensitivity to the antiviral activity.

“…Inhibition of Cell Proliferation. Daudi cells are extremely sensitive to the antiproliferative effects of human IFN-a or -,B (1)(2)(3)(4)(5). Treatment of the cells with 10 units/ml for 24 hr produced a small decrease in cell number (=18%), and by 48 *hr the inhibition of growth was very marked (Fig.…”

“…The Burkitt lymphoma-derived human lymphoblastoid cell line Daudi is particularly sensitive to the antiproliferative effects of human interferons (1)(2)(3), and inhibition of cell proliferation is accompanied by a large decrease in the ability of the cells to incorporate exogenous [3H]thymidine into DNA (3)(4)(5). The inhibition of cell growth after treatment with naturally occurring or recombinant interferon a (IFN-a) is associated with the differentiation of Daudi cells from an immature B cell phenotype into cells with several characteristics of mature plasma cells (6).…”

“…In addition, participation of IDO in host-cell defense mechanisms against proliferation of intracellular parasites following IFN-y treatment has suggested another important biological role for this enzyme (34,35). In light of the known antiproliferative actions of IFNs on either normal or malignant cells (36)(37)(38)(39)(40)(41)(42)(43)(44) and of the requirement of L-tryptophan, the least abundant amino acid, for protein synthesis and cellular growth (45)(46)(47)(48), we investigated whether the induction of IDO constitutes another mechanism to explain the antiproliferative action of IFNs.…”

“…Even though P3HR-1 and Ag-876 expressing EBNA and EA/VCA, and Raji cells expressing only EBNA were not affected by the treatment with the LDV/7 interferon, these virusinfected cells could be destroyed by effector cells activated by interferon (16,17). Since the inhibition of transformation of cord blood cells by Ag-876 EBV was observed in low multiplicity of infection, perhaps more purified and higher doses of interferon may prevent transformation even with higher levels of viruses.…”
