The purpose of this study was to determine the motility and viability of kampung rooster (Gallus gallus domesticus) spermatozoa in 0.9% Sodium chloride (NaCl) solution as an extender with the addition of egg yolk at different concentrations at cool temperature (5°C). This study was performed using two 1.5 years old healthy local roosters. Semen was collected through massage in the morning. The pooled semen sample was divided into four treatment groups. In the T0 group the semen was diluted in 0.9% NaCl, while in the T1, T2 and T3 groups the semen was diluted in 0.9% NaCl added with 5, 10 and 15% egg yolk. The results showed that the motility and viability of spermatozoa decreased when stored at cool temperatures for ten hours (p <0.05). Semen of roosters stored at 5°C in saline solution without the addition of egg yolks showed the lowest motility and viability of spermatozoa (p <0.05). The addition of egg yolk into the saline extender increased the motility and viability of spermatozoa. Concentration of 15% egg yolk in saline solution resulted in the highest spermatozoa motility and viability when stored for up to 8 hours (p <0.05). However, motility and viability of spermatozoa at 10 hours of storage were not significantly different (p >0.05) with the addition of 10% and 15% egg yolks. Therefore, it could be concluded that the addition of 15% egg yolk into a saline solution as an extender could maintain the motility and viability of kampung rooster spermatozoa when stored at 5°C for 10 hours.

Background and Aim: The management of incubation conditions impacts embryonic development, hatchability, and post-hatch performance. This study aimed to examine the effects of thermal manipulation (TM) during embryonic development on roosters’ thermotolerance, antioxidant activity, immunity, and semen quality under heat-stress conditions. Materials and Methods: 1200 fertile eggs were distributed evenly between two groups, each containing three replicates (200 eggs/replicate). The first group (G1) was held in the commercial setter with a consistent temperature of 37.5°C and 55% relative humidity (RH) through the 18-day incubation period, acting as a control, while the second group (G2) experienced these conditions until only the 11th day. The eggs were incubated at 39.5°C with 60% RH for 4 h each day from the 12th to the 18th day. From the 19th to 22nd incubation days, both groups maintained a consistent temperature of 37.2°C with a RH of 70%. Two hundred hatched male chicks per treatment group were moved into a closed-system house. All roosters were exposed to a 6-h daily heat challenge with a temperature of 35°C and a humidity of 70% between their 36th and 40th weeks of age. Results: Roosters of G2 exposed to thermal challenge showed improvements (p ≤ 0.05) in multiple blood biochemical, antioxidant, and immunity markers, including total protein, globulin, aspartate aminotransferase, alanine aminotransferase, triiodothyronine, thyroxine, corticosterone, testosterone, total antioxidant capacity, malondialdehyde, immunoglobulin G, immunoglobulin M, and immunoglobulin A levels. Improved semen quality characteristics, including ejaculate volume, sperm concentration, motility, livability, and quality factor, as well as enhanced thermoregulation in post-hatch cocks, were also achieved (p ≤ 0.05). Conclusion: To boost antioxidant activity, immunity, thermotolerance, and semen parameters in roosters under heat-stress conditions, TM application during egg incubation, specifically at 12–18 days, is recommended. In a 10-wk study, alterations in the rate of fertility, egg viability, and hatch parameters of adult geese exposed to different breeding methods were investigated. Twenty-four matured geese (4.0 ± 0.45 average weight) were randomly divided into three groups (TNM—natural mating group, TIM—artificial insemination group, TNI—natural mating and insemination group) of two replicates with four geese per replicate in a completely randomized design. Fresh semen collected from six ganders (5.2 ± 0.69 average weight) was pooled and used to inseminate the geese in TIM and TN1 at 0.2Ml at insemination times. The geese in TNM and TNI were allowed to mate naturally. Insemination and mating was done at 3 d interval and eggs from each treatment were collected daily. Incubation of eggs was done weekly, candling and transfer to hatcher were done on day 27 and goslings hatched out on day 30. Fertility, early embryo mortality (EEM), mid embryo mortality (MEM), late embryo mortality (LEM), hatch of fertile eggs (HOF), and hatch of set eggs (HOS) were obtained and analyzed using descriptive statistics and ANOVA and means separated using least significant difference test. Geese in TNI had significantly higher fertility (93.33 ± 10.97%) than TNM (59.67 ± 31.29%) and TIM (83.60 ± 17.14%). The EEM was higher in TIM than in the two other groups while the HOF and HOS were higher in TNM and TNI than in TIM. This study suggests that in comparison with TIM, higher fertility, hatchability, and lower embryo mortality can be obtained when geese are inseminated and naturally mated simultaneously.
