Self‐assembled Cupric Oxide Nanoclusters for Highly efficient chemodynamic therapy

Abstract: Chemodynamic therapy (CDT) based on Fenton and Fenton-like reactions induces cancer cell killing via in situ catalyzing H 2 O 2 and generating highly oxidative hydroxyl radicals ( * OH) in tumor sites. Their application is not limited by tumor grown depth or hypoxic microenvironment. However, the reaction efficiency is still hampered due to the structure of catalytic agents and the requirement for low pH environment. Here, we design a porous CuO nanocluster (CuO NC) through self-assembly of oleylamine stabiliz… Show more

“…Released Cu 2+ catalyzed over-expressed H 2 O 2 intra-bladder cancer cells into • OH via the Fenton-like reaction, and induced cell apoptosis. To evaluate intracellular • OH generation, DCFH-DA, a fluorescent probe which is oxidized by ROS to generate DCF with green fluorescence, 44 was incubated with LipGel-NMs treated MB49 cells, which showed intense intracellular DCF fluorescence, whereas negligible intracellular DCF fluorescence was observed for MB49 cells that treated with NC-LipGel-NMs that did not contain CuO 2 (Fig. 3A, LipGel-NMs, NC-LipGel-NMs).…”

“…Subsequently, they were modified with folic acid (FA) to target cancer cells, and the obtained CuO NCs-FA were delivered to HeLa cells to verify the feasibility of intracellular Fenton-like reactions. 136 In addition, polydopamine-modified CuO combined with natural glucose oxidase (GOD), in which CuO maximizes the cascade catalytic efficiency with a high loading rate (47.1%) due to the elaborate hollow mesoporous structure (Fig. 10b), was combined with NIR-II-induced thermal effects for combined O 2 -promoted starvation and photothermal-chemical kinetic treatment.…”

“…Specifically, CuO is commonly used in CDT to induce intracellular ·OH generation for tumor cell eradication. For example, Jin et al successfully self-assembled CuO nanoparticles (NPs) with an average size of 5 nm into CuO nanospheres (Figure a) through a surfactant-assisted method . The as-obtained CuO nanospheres showed a porous structure that provided a significantly large specific surface area (395.5 m 2 g –1 ), which enhanced active reaction sites and improved Fenton-like reaction efficiency.…”
