Selenium Supplementation Reduced Oxidative DNA Damage in Adnexectomized BRCA1 Mutations Carriers

Abstract: Some experimental evidence suggests that BRCA1 plays a role in repair of oxidative DNA damage. Selenium has anticancer properties that are linked with protection against oxidative stress. To assess whether supplementation of BRCA1 mutation carriers with selenium have a beneficial effect concerning oxidative stress/DNA damage in the present double-blinded placebo control study, we determined 8-oxodG level in cellular DNA and urinary excretion of 8-oxodG and 8-oxoGua in the mutation carriers. We found that 8-oxo… Show more

“…We assessed the role of drugs used in patients with septic shock that promote vasoconstriction (norepinephrine, AVP) (46), of a stabilized analogue of AVP triggering the release of VWF from endothelial cells (DDAVP) (47), selenite supplementation with known antioxidative properties (48) and APC which is reported to regulate severe inflammatory response (49). In HSCs, all tested compounds displayed nonsignificant effects on ADAMTS13 transcription at 6 h ( Figure 2A).…”

“…Therefore, increase in urinary excretion of the modified base in the group with longer life expectancy is, most likely, a measure of better response to the radiation (higher radiosensitivity) and simultaneously efficient removal of the damage from cellular DNA of this group (14). Because 8-oxo-Gua is a repair product of the DNA damage, it is likely that, at least in the case of some patients with the higher activity of OGG1 the combination of higher OGG1 repair capacity and irradiation was associated with stable background level of 8-oxo-Gua in cellular DNA.…”

“…Zinc is an essential cofactor of various metalloenzymes and it protects the skin from UV irradiation and has been used as a destructive agent for the management of cancer [7,8]. Sodium selenate enhances the repair system of DNA segments and reduces the risk of new cancer development [9,10]. Numerous literature suggests that it prevents skin cancers in combination with L-selenomethionine [11,12].…”
