Selective α1A-blocker improves bladder storage function in rats via suppression of C-fiber afferent activity

Abstract: These results suggest that the α(1A)-AR subtype activates C-fiber afferents, and that consequently α(1A)-blockade can improve bladder storage function.

“…It has been shown that suppression of afferent nerve activity with neurotoxins, such as botulinum toxin and resiniferatoxin, effectively relieves OAB symptoms in both human patients (Schurch et al 2000;Silva et al 2002) and animal models (Atiemo et al 2005;Khera et al 2005;Yokoyama et al 2005Yokoyama et al , 2010, emphasizing the importance of afferent nerve dynamics in the pathogenesis of OAB.…”

“…Oyama et al. more recently also demonstrated better improvements in IPSS score with silodosin vs. tamsulosin or naftopidil up to 9 months after BT 95. Shimizu et al., however, demonstrated that the effects of silodosin are temporary in a 12‐month follow‐up study of 105 patients given sildonosin daily for 6 months immediately after BT 96.…”

“…Finally, this binding induces an increase in the intracellular calcium levels and smooth muscle contraction [15]. The main cause of LUTS in BPH is prostate contraction that is predominantly mediated by alpha1A-AR [16,17]. Therefore, the blockage of α1A adrenoreceptors results in prostatic and urethral smooth muscle relaxation and may improve the voiding symptoms.…”
