Selective uptake of naked vaccine RNA by dendritic cells is driven by macropinocytosis and abrogated upon DC maturation

Abstract: Even though it is known for more than one decade that antigen-encoding RNA can deliver antigenic information to induce antigen-specific immunity against cancer, the nature and mechanism of RNA uptake have remained enigmatic. In this study, we investigated the pharmacokinetics of naked RNA administered into the lymph node. We observed that RNA is rapidly and selectively uptaken by lymph node dendritic cells (DCs). Furthermore, in vitro and in vivo studies revealed that the efficient internalization of RNA by hu… Show more

“…In our approach, however, CD8a þ TiDCs are the target, and these cells were shown to engulf and translate mRNA dissolved in Hartmann solution. Most likely, the mRNA uptake is mediated by macropinocytosis as described for the mRNA uptake by intranodal DCs (38). Consequently, when targeting DCs in situ, mRNA is an attractive vector as it enables selective delivery without prior manipulation.…”

“…RNA delivers the complete antigen, is selectively internalized by professional antigen-presenting cells (3), and has an intrinsic adjuvant activity. RNA does not integrate into the genome and with the transient expression of the encoded antigens allows better control of immune responses.…”

“…An exception to this are immature dendritic cells, which efficiently take up and accumulate mRNA by macropinocytosis [69]. By contrast, effective delivery of mRNA in other cell types requires alternative delivery methods.…”
