A cultura do trigo (Triticum aestivum L.) está sujeita ao ataque de vários patógenos causadores de manchas foliares que, dependendo da intensidade podem causar reduções significativas no rendimento de grãos da cultura (12).A mancha-amarela do trigo encontra-se distribuída em mais de 21 países (10) sendo considerada uma doença cosmopolita. Foi relatada como doença importante na Austrália (13), no Canadá (20) e nos Estados Unidos (8). No Brasil, a primeira referência sobre o agente causal foi no Rio Grande do Sul, em 1959 (4). A partir do ano de 1979, a mancha-amarela aumentou de intensidade, especialmente em função da semeadura direta, e atualmente é uma das doenças foliares mais frequentes em trigo (10). RESUMOPalavras-chave adicionais: Triticum aestivum, mancha-amarela, esporos, regimes de luz A mancha-amarela do trigo, causada pelo fungo Drechslera tritici-repentis, é de ocorrência mundial e uma das principais doenças da cultura. No Brasil, a mesma está presente em todas as safras e pode reduzir o rendimento de grãos em mais de 40%. A sua ampla adaptabilidade a ambientes diversos pode estar relacionada à capacidade dos esporos germinarem sob diferentes temperaturas, assunto que foi examinado neste experimento conduzido na UPF, em 2011. Suspensões de conídios (350 μL) foram depositadas em placas de Petri com meio ágar-água e incubadas em câmara tipo BOD, na presença e ausência de luz, a -5, 0, 5, 10, 15, 20, 25, 30, 35 Seu agente causal, o fungo Drechslera tritici-repentis Died., sobrevive, principalmente, em sementes e restos culturais, a partir dos quais infecta as folhas causando lesões primárias. Nessas manchas o fungo produz novos esporos, disseminados pelo vento até as folhas ou plantas vizinhas, originando ciclos secundários da doença (17).As condições ambientais ideais para o desenvolvimento da manchaamarela incluem temperaturas entre 18 e 28ºC e período de molhamento igual ou superior a 30h (15). A temperatura age como catalisador dos processos biológicos, razão pela qual tanto as plantas como os patógenos requerem uma temperatura mínima e máxima para crescer e desenvolver normalmente suas atividades (16).Apesar da importância da doença, a literatura sobre aspectos da Tan spot, caused by the fungus Drechslera tritici-repentis, is of worldwide occurrence and one of them main diseases affecting the wheat culture. In Brazil, tan spot is present in every crop season and may reduce grain yield by more than 40%. The great adaptability of this disease to diverse environments may be related to the capability of spores to germinate at different temperatures, which was examined in this experiment conducted at UPF, in 2011. Northern corn leaf blight, caused by Exserohilum turcicum (Et), is a disease of widespread occurrence in regions where corn, sweetcorn and popcorn are grown. This disease has great potential to cause damage and has been studied for years, but the association of its causal agent with seeds remains unconfirmed. Thus, the availability of a sensitive method to detect and quantify the inoculum in seeds, even at low incidence, is essential. The aim of this study was to develop a method to detect and quantify the presence of the fungus infecting and infesting corn and popcorn seeds. Artificially and naturally infected seeds were employed to develop the medium. ABSTRACT(60 mg/L), captan (30 mg/L), streptomycin sulfate (500 mg/L) and neomycin sulfate (600 mg/L) aggregated to the medium lactose casein hydrolysate agar medium. By using this, Et was detected in naturally infected corn seeds, showing 0.124% incidence, in four out of ten analyzed samples. In addition, 1.04 conidia were detected per infested seed. By means of isolation, pathogenicity test, morphological characterization and comparison with descriptions of the species in the literature, the fungus isolated from the seeds was confirmed to be Et. Both infection and infestation were considered low; thus, for studies of Et detection in corn seeds, the use of semi-selective medium and more than 1,200 seeds/sample is suggested.

Several fungal species belonging to the genus Fusarium cause agriculturally important diseases in a variety of plants. The pathogen Fusarium graminearum Schwabe, teleomorph Gibberella zeae (Schwein.) Petch, is the causal agent of wheat (Triticum aestivum L.) head blight also known as giberela ear rot and maize (Zea mays L.) stem and cob rot (2). This parasite has a wide geographical distribution and attacks a large number of crops causing qualitative and quantitative damage, which can affect agricultural production (6). Head blight of wheat is particularly important in the state of Rio Grande do Sul, Brazil, where wheat is extensively grown (13). In the south of Brazil ecological group II occurs (1, 11). A detailed knowledge of the survival and dissemination mechanisms of pathogenic fungi is important in developing control measures for difficult-to-control fungal plant pathogens. Selective or semi-selective culture media, which act by excluding non-target fungi and bacteria, are important tools for fungal isolation, purification and quantification and for investigating the detailed biology of fungi, being especially useful for slow growing organisms (3, 4,12). However, the media currently available for Fusarium do not meet criteria such as sensitivity and selectivity and ability to allow the development of Fusarium colonies with consistent morphological characteristics and pigmentation (9).Several selective or semi-selective culture media have been developed for the isolation of Fusarium species, including the most widely-used culture medium Nash-Snyder agar (NSA) (7). However, the NSA medium often does not maintain the ABSTRACTFungi of the genus Fusarium cause a variety of difficult to control diseases in different crops, including winter cereals and maize. Among the species of this genus Fusarium graminearum deserves attention. The aim of this work was to develop a semi-selective medium to study this fungus. In several experiments, substrates for fungal growth were tested, including fungicides and antibiotics such as iprodiona, nystatin and triadimenol, and the antibacterial agents streptomycin and neomycin sulfate. Five seed samples of wheat, barley, oat, black beans and soybeans for Keywords: Small grains scab, head blight, Gibberella zeae, seed assay.quarter dextrose agar (1/4PDA; potato 50g; dextrose 5g and agar 20g ), either un supplemen ted or supplemented with various concentrations of the antimicrobial agents cited above. The selected components and concentrations (g.L -1 ) of the proposed medium, Segalin & Reis agar (SRA-FG), were: iprodiona 0.05; nystatin 0,025; triadimenol 0.015; neomycin sulfate 0.05; and streptomycin sulfate, 0.3 added of ¼ potato sucrose agar. In the isolation from seeds of cited plant species, the sensitivity of this medium was similar to that of NSA but with de advantage of maintaining the colony morphological aspects similar to those observed in potato-dextrose-agar medium.
